为了解2012-2014年福建省流行 PEDV 毒株 E 和 M 基因的变异情况,对19株 PEDV 的 E 基因和18株 PEDV 的 M 基因进行克隆和序列分析。结果表明:与 CV777标准毒株相比较,福建流行 PEDV 毒株的 E 和M 基因存在编码的部分氨基酸变异。19株 PEDV E 基因的核苷酸之间的同源性为97.4%~100.0%,与attenuated DR13弱毒株和 CV777标准株同源性分别为97.4%~100.0%和97.0%~99.6%。18株 PEDV M 基因的核苷酸之间的同源性为97.4%~100.0%,与 attenuated DR13弱毒株和 CV777标准株同源性分别为97.2%~99.6%和97.5%~98.4%。核苷酸遗传进化树表明福建流行 PEDV 毒株的 E 基因与 CV777亲缘关系较近,而M 基因与2008年泰国流行的 KU05CB08毒株亲缘关系较近,是个新的基因型,可能源于泰国。

To investigate the proteinvariationsin the porcine epidemic diarrhea virus (PEDV),19 E and 18 M genes from the viruses collected from different farms in Fujian in 2012 -2014 were cloned and sequenced.The results indicated that the PEDV E and M genes were conservative.There were individual variants foundonlyonamino acids, as compared with CV777 strain.On the nucleotide sequences,the E geneswere 97.4% - 100.0% homogenous among themselves,97.4%-100.0% homogenous with attenuated DR13 strain,and 97.0%-99.6% homogenous with CV777 strain.For the M genes,they were 97.4%-100.0% homogenous among the 18 of them,97.2% -99.6% homogenous with attenuated DR13,and 97.5%-98.4% homogenous with CV777. A phylogenetic analysis indicated that the E genes of the 19 PEDVs were closely related to CV777,and the M genes of the 18 PEDVs closely related to KU05CB08.The M gene wasa new variant,which might bear an origin of strains from Thailand.
