[目的]研究T RPV1在SD大鼠一过性肠道感染旋毛虫后形成内脏高敏感中的作用。[方法]30只大鼠随机分为正常对照组、假手术组和内脏高敏感组。内脏高敏感组给予旋毛虫感染,假手术组给予同等量的 PBS灌胃,正常对照组不作任何处理。8周后,观察各组结直肠不同扩张压力下(20 mmHg、40 mmHg、60 mmHg、80 mm‐Hg)腹壁肌电活动变化;采用Western‐blot检测远端结肠TRPV1蛋白质水平的表达。[结果]①正常对照组、假手术组和内脏高敏感组结直肠不同扩张压力与腹壁肌电活动之间有显著相关性,各组相关系数与 P值分别为:正常对照组 r=0.825、P=0.000,假手术组 r=0.831、P=0.000,内脏高敏感组 r=0.625、P=0.006。在20 m m H g、40 mmHg和60 mmHg扩张压力下,3组间AUC均差异有统计学意义(P<0.05);在80 mmHg ,3组间AUC差异无统计学意义(P>0.05)。②正常对照组、假手术组、内脏高敏感组 TRPV1光密度相对值分别为0.95±0.17、0.94±0.24、1.57±0.31,内脏高敏感组较正常对照组、假手术组均显著升高( P<0.05)。[结论]内脏高敏感的形成与T RPV1高表达密切相关。

[목적]연구T RPV1재SD대서일과성장도감염선모충후형성내장고민감중적작용。[방법]30지대서수궤분위정상대조조、가수술조화내장고민감조。내장고민감조급여선모충감염,가수술조급여동등량적 PBS관위,정상대조조불작임하처리。8주후,관찰각조결직장불동확장압력하(20 mmHg、40 mmHg、60 mmHg、80 mm‐Hg)복벽기전활동변화;채용Western‐blot검측원단결장TRPV1단백질수평적표체。[결과]①정상대조조、가수술조화내장고민감조결직장불동확장압력여복벽기전활동지간유현저상관성,각조상관계수여 P치분별위:정상대조조 r=0.825、P=0.000,가수술조 r=0.831、P=0.000,내장고민감조 r=0.625、P=0.006。재20 m m H g、40 mmHg화60 mmHg확장압력하,3조간AUC균차이유통계학의의(P<0.05);재80 mmHg ,3조간AUC차이무통계학의의(P>0.05)。②정상대조조、가수술조、내장고민감조 TRPV1광밀도상대치분별위0.95±0.17、0.94±0.24、1.57±0.31,내장고민감조교정상대조조、가수술조균현저승고( P<0.05)。[결론]내장고민감적형성여T RPV1고표체밀절상관。
Objective]To investigate the role of TRPV1 on the formation of visceral hypersensitivity on rats induced by transient intestinal infection .[Methods]Thirty male Sprague‐Dawley rats were randomly divided into normal control ,sham operation and visceral hypersensitivity .The rats in visceral hypersensi‐tivity were infected by administering 1 .0 ml of PBS containing 8 000 T .spiralis larvae by gavage .Those rats in sham operation were only administered 1 .0 ml of PBS .Electromyography (EMG) was recorded at 8th week after adaptive feeding for evaluating visceral sensitivity .The areas under curve (AUC) of EMG in 20 s were calculated to evaluate the visceral sensitivity .Western‐blot was respectively employed to de‐tect protein TRPV1 expression .[Results]① Under different grades levels of CRD pressure ,AUCs showed significant correlation in the three groups respectively (P<0 .05) .The coefficient of correlation and P val‐ue of the three groups were 0 .825 ,P=0 .000;0 .831 ,0 .000 ;0 .625 ,0 .006 respectively .Under 20 mmHg , 40mmHg and 60mmHg ,the difference of the AUC among the three groups was significant ( P=0 .034 , 0.043 ,0 .048 respectively) .Under 80 mmHg ,no significant difference was found in three groups (P>0.05) .②In normal control ,sham operation and visceral hypersensitivity group ,the IODs of TRPV1 pro‐tein were 0 .95 ± 0 .17 ,0 .937 ± 0 .24 ,1 .57 ± 0 .31 respectively .The expression of TRPV1 of visceral hyper‐sensitivity was significantly increased than sham operation and normal control (P< 0 .05) .[Conclusion] The increased expression of TRPV1 plays an important role in the formation of visceral hypersensitivity .
