目的:建立反相高效液相色谱法测定冻干粉针中 DHEA 含量。方法:采用 Diamonsil C18(250 mm×4.6 mm,5μm)色谱柱,流动相为乙腈-水(55:45),流速1.0 mL · min-1;检测波长206 nm ;进样量20μL ;柱温:室温。结果:在此色谱条件下 DHEA 与有关物质能达到较好分离,在0.01 mg · mL-1~0.7 mg · mL-1浓度范围内与峰面积呈良好线性关系,r=0.9998;方法重复性 RSD 为0.42%;加样平均回收率(n=6)为98.91%。结论:该方法准确可靠,灵敏度高,专属性好,可用于测定 DHEA 冻干粉针含量。

Objective: To establish an RP-HPLC method for determination of the content of DHEA. Methods:Diamonsil C18 (250 mm× 4.6 mm,5 μm) column was adopted ; the mobile phase consisted of a mixture of acetonitrile - water (55:45) at the flow rate was 1.0 mL·min- 1 ;The ultraviolet detection wavelength was set at 206 nm; Loaded volume was 20 μL; the column temperature was room temperature. Results: DHEA and its related substances could be separated well. The linearity of DHEA was well correlated ( r=0.9998) within the range of 0.01 mg · mL- 1~0.7 mg · mL- 1. The method precision RSD was 0.42%. The average recoveries (n=6) was 98.91%. Conclusion: The method appeared to be accurate, reliable, sensitive, specific and can be used for the determination of the content of freeze-dried powder injection DHEA .
