Vitamin D3%Porphyromonas gingvialis(Pg)%immune response%periodontitis model
目的:利用小鼠实验性皮下小室牙周炎模型,研究维生素D3(VD3)对牙龈卟啉单胞菌(Porphyromonas gingvialis,Pg)诱导的适应性免疫的作用。方法:选用15只8~10周龄BALB/c雌性小鼠,以口腔用不锈钢丝弯制簧圈小室,埋于小鼠两侧背部皮下建立小室模型。创口愈合后将小鼠分成VD3组、无水乙醇组(溶剂对照组)及磷酸盐缓冲液组(空白对照组),通过腹腔注射连续给药14 d。第7 d和第14 d,分别向皮下小室内注射Pg菌液造成局部感染。收集感染前和第二次感染后第7 d的血清样本,酶联免疫吸附实验(ELISA)检测Pg特异性的抗体水平。取第二次感染后第7 d的小鼠脾脏细胞,经Pg再次刺激,收集细胞培养上清,ELISA分析上清中的Th1、Th2、Th17细胞因子水平。结果:与无水乙醇组相比,VD3处理组的脾细胞产生了较低水平的促炎细胞因子,产生了较高水平的抗炎细胞因子,其中IFN-γ和IL-10在两组之间有统计学差异(P<0.05)。Pg特异性的IgG,IgG1和IgG 2a在两组之间无显著性差异。结论:VD3可以调节宿主对Pg的适应性免疫反应,在调节炎性细胞和控制炎症发展方面起着重要的作用。

목적:이용소서실험성피하소실아주염모형,연구유생소D3(VD3)대아간계람단포균(Porphyromonas gingvialis,Pg)유도적괄응성면역적작용。방법:선용15지8~10주령BALB/c자성소서,이구강용불수강사만제황권소실,매우소서량측배부피하건립소실모형。창구유합후장소서분성VD3조、무수을순조(용제대조조)급린산염완충액조(공백대조조),통과복강주사련속급약14 d。제7 d화제14 d,분별향피하소실내주사Pg균액조성국부감염。수집감염전화제이차감염후제7 d적혈청양본,매련면역흡부실험(ELISA)검측Pg특이성적항체수평。취제이차감염후제7 d적소서비장세포,경Pg재차자격,수집세포배양상청,ELISA분석상청중적Th1、Th2、Th17세포인자수평。결과:여무수을순조상비,VD3처리조적비세포산생료교저수평적촉염세포인자,산생료교고수평적항염세포인자,기중IFN-γ화IL-10재량조지간유통계학차이(P<0.05)。Pg특이성적IgG,IgG1화IgG 2a재량조지간무현저성차이。결론:VD3가이조절숙주대Pg적괄응성면역반응,재조절염성세포화공제염증발전방면기착중요적작용。
Objective:To explore the effects of vitamin D3(VD3) on the adaptive immune responses in mice induced by porphyromonas gingvialis (Pg) by using a subcutaneous chamber model. Method:Fifteen BALB/c female mice aged 8~10 weeks were used in this study. Stainless steel coils of 10 mm in length and 5 mm in diameter were made. Two coils were separately implanted in the lateral dorsolumbar regions of each mouse to create subcutaneous chambers. After the wound healed,animals were intraperitoneally injected with VD3(VD3 group), absolute ethanol(dissolvant control group) or Phos-phate buffer saline (PBS group) for 14 days. On day 7 and day 14,each chamber was infected with 1×109 CFU Pg. Serum samples were collected before and after Pg infection for antibody detection by enzyme-linked immunosorbent assay(ELISA). On day 21,spleens were harvested and splenocyte suspensions were prepared and stimulated with Pg. Cell supernatants were then collected for cytokines level assessment by ELISA. Result:The splenocytes in VD3 group produced lower levels of pro-inflammatory cytokines and higher levels of anti-inflammatory cytokines compared to AE group. There exist significant differ-ence in IFN-γand IL-10 between the two groups. Serum antibody levels didn't show significant differences between the two groups. Conclusion:VD3 can modulate the adaptive immune responses to Pg and play an important role in regulating in-flammatory cells and controling the development of inflammation.
