目的:利用静电纺丝法制备载盐酸多西环素(DOX)纳米纤维膜片,评价其作为局部药物治疗与GTR/GBR技术的结合,在治疗种植体周围炎方面的可行性的相关基础研究。方法:用静电纺丝法制备载DOX的聚己内酯(PCL)纳米纤维膜。扫描电子显微镜观察纤维膜的表面形貌、分析药物对纤维形貌的影响,差示量热扫描法(DSC)分析药物在纤维中的分布状态,琼脂扩散实验检测载药膜片对牙龈卟啉单胞菌(Pg)和伴放线放线杆菌(Aa)的抑菌作用。结果:制备的DOX-PCL纳米纤维表面光滑,直径分布均匀,随着DOX的载药量由15%增加到25%,纤维直径由247.17 nm降低到181.34 nm;同时以无定形分子形式分布于纳米纤维中的DOX对Aa和Pg均具有明显的抑菌作用。结论:作为药物治疗与GTR/GBR技术的结合,静电纺丝法制备的载药GTR/GBR膜在种植体周围炎的治疗方面具有良好的应用前景。

Objective:To prepare a doxycycline hydrochloride(DOX)-loaded nanofibrous membrane by electrospining method. Evaluate its relative foundational properties for peri-implantitis therapy as the combination of local drug treatment and GTR/GBR surgery. Method:Different groups' DOX-loaded PCL nanofibrous membranes were prepared by eletrospun. Then we observed the morphology and the diameter distribution of nanofibers,investigated the manner of DOX in drug-load-ed nanofibers and its bioactivity against Aggregatibacter actinomycetemcomitans (Aa) and Porphyromonas gingivalis(Pg). Result:DOX-loaded PCL nanofibers were uniform distributed and the surface of fibers were smooth. Fibrous average diame-ters decreased from 247.16 nm to 181.34nm with the drug proportion increasing from 15%to 25%w/w. Differential scan-ning calorimetry (DSC) test showed DOX appeared an amorphous manner in the polymer matrix. And DOX-loaded mem-branes had an obvious inhibited effect on Aggregatibacter actinomycetemcomitans (Aa) and Porphyromonas gingivalis(Pg), respectively. Conclusion:As the combination of local drug treatment and GTR/GBR surgery,electrospinning DOX-loaded PCL nanofibrous membrane has a well prospect on the field of peri-implantitis therapy.
