目的:观察荧光标记物 DiI 在已固定的人胚胎周围神经中的示踪显影,探讨 DiI 在周围神经示踪中的应用可行性。方法利用人胚胎新鲜标本(非疾病死亡的引产胎儿),胎龄>20 w,经升主动脉灌注10%福尔马林进行固定,并浸泡于固定液8~10 d 后,将微量的 DiI 结晶分别植入人胚胎标本三叉神经的眼神经、上颌神经、下颌神经及三叉神经根中,放入固定液在37℃恒温箱内贮存3个月,经冰冻切片,置荧光显微镜下观察评价示踪效果。结果DiI 注入眼神经、上颌神经及下颌神经中后,可在三叉神经节内观察到许多逆行标记的神经节细胞和纤维,且细胞及细胞突起显示较好;DiI 注入三叉神经根中后,三叉神经节内也可见许多顺行标记的神经节细胞和纤维,细胞及细胞突起也显示较好。结论DiI 是一种良好的荧光类双向神经示踪剂,用于已固定人胚胎周围神经束路的失踪研究是可行的。
目的:觀察熒光標記物 DiI 在已固定的人胚胎週圍神經中的示蹤顯影,探討 DiI 在週圍神經示蹤中的應用可行性。方法利用人胚胎新鮮標本(非疾病死亡的引產胎兒),胎齡>20 w,經升主動脈灌註10%福爾馬林進行固定,併浸泡于固定液8~10 d 後,將微量的 DiI 結晶分彆植入人胚胎標本三扠神經的眼神經、上頜神經、下頜神經及三扠神經根中,放入固定液在37℃恆溫箱內貯存3箇月,經冰凍切片,置熒光顯微鏡下觀察評價示蹤效果。結果DiI 註入眼神經、上頜神經及下頜神經中後,可在三扠神經節內觀察到許多逆行標記的神經節細胞和纖維,且細胞及細胞突起顯示較好;DiI 註入三扠神經根中後,三扠神經節內也可見許多順行標記的神經節細胞和纖維,細胞及細胞突起也顯示較好。結論DiI 是一種良好的熒光類雙嚮神經示蹤劑,用于已固定人胚胎週圍神經束路的失蹤研究是可行的。
목적:관찰형광표기물 DiI 재이고정적인배태주위신경중적시종현영,탐토 DiI 재주위신경시종중적응용가행성。방법이용인배태신선표본(비질병사망적인산태인),태령>20 w,경승주동맥관주10%복이마림진행고정,병침포우고정액8~10 d 후,장미량적 DiI 결정분별식입인배태표본삼차신경적안신경、상합신경、하합신경급삼차신경근중,방입고정액재37℃항온상내저존3개월,경빙동절편,치형광현미경하관찰평개시종효과。결과DiI 주입안신경、상합신경급하합신경중후,가재삼차신경절내관찰도허다역행표기적신경절세포화섬유,차세포급세포돌기현시교호;DiI 주입삼차신경근중후,삼차신경절내야가견허다순행표기적신경절세포화섬유,세포급세포돌기야현시교호。결론DiI 시일충량호적형광류쌍향신경시종제,용우이고정인배태주위신경속로적실종연구시가행적。
Objective To observe the fluorescent marker DiI which has been fixed in the peripheral nerves of human embryo,investigate the feasibility of the tracing of the DiI in the human peripheral nerves.Methods Use the hu-man embryonic specimens (non-disease induction of fetal deaths,more than 20 weeks gestational age),after as-cending aorta perfusion fixed in 10% formalin and immersed in the same fixative eight to ten days,will trace the DiI crystalline specimens were implanted through the ophthalmic nerve,maxillary nerve,mandibular nerve of the trigeminal nerve and the trigeminal nerve root of human embryo,storage incubator at 37 degrees for two to three months,Frozen sections;Evaluation the effect of tracer by fluorescence microscope Results The experimental group,which is DiI injected in the ophthalmic nerve,maxillary nerve,mandibular nerve of the trigeminal nerve can observe many retrograde labeling of ganglion cells and fibers,also observe well labeled cells and processes;in the root group also observed that many Anterograde labeled ganglion cells and fibers,and the cells and cell proces-ses also showed clearly.Conclusion DiI is a best two-way neural tracers,it is feasible and can be used to trace the peripheral nerves in fixed human embryo.
