当前吸毒者吸食的毒品主要是海洛因。海洛因在体内代谢很快,半衰期仅为2.0 min。海洛因在体内首先代谢成单乙酰吗啡、而后代谢成吗啡和葡萄糖醛酸吗啡,因此在检材中检出单乙酰吗啡就能证明吸毒者吸食的为海洛因。单乙酰吗啡主要通过尿液排泄,血液中的含量很小,难于检测。目前单乙酰吗啡的检测方法国内外已有不少报道,有气相色谱法、高效液相色谱法、气-质联用法等,然而用这些方法均不能很好的检验样品的中的单乙酰吗啡。本文采用固相萃取法提取,通过调pH值看回收率,确定最佳pH值,最大程度地减少了样品处理过程对单乙酰吗啡含量的影响。同时本方法采用HPLC-MS/MS检验,降低了噪音,提高了灵敏度。
