目的:研究右美托咪定与维生素C单独或联合使用对小鼠小肠缺血再灌注损伤( I/R)的保护作用及可能机制。方法雄性小鼠50只,随机分为5组( n=10):假手术组( sham组,暴露肠系膜上动脉,不结扎),肠缺血再灌注组(I/R组),右美托咪定组(DEX组,提前30 min注射DEX),维生素C组(VC组,提前10 min注射VC),右美托咪定联合维生素C组( DEX+VC组,提前30 min注射DEX,提前10 min注射VC)。缺血20 min再灌注24 h后取小肠组织,苏木精-伊红染色观察小肠黏膜的损伤情况,免疫组化法检测小肠黏膜中白细胞介素-6(IL-6)、肿瘤坏死因子α(TNF-α)和caspase-3的表达变化并做chiu′s评分。结果 DEX、VC、DEX+VC组小肠黏膜的chiu′s评分和IL-6、TNF-α、caspase-3的表达较I/R组降低(P<0.05),而较sham组升高(P<0.05);DEX组和VC组差异无统计学意义( P>0.05),DEX +VC组小肠黏膜的chiu′s评分和IL-6、TNF-α、caspase-3的表达较DEX、VC组降低(P<0.05)。结论右美托咪定、维生素C对小鼠小肠缺血再灌注损伤有保护作用,两者联用有协同作用,且保护作用与IL-6,TNF-α和caspase-3有关。

목적:연구우미탁미정여유생소C단독혹연합사용대소서소장결혈재관주손상( I/R)적보호작용급가능궤제。방법웅성소서50지,수궤분위5조( n=10):가수술조( sham조,폭로장계막상동맥,불결찰),장결혈재관주조(I/R조),우미탁미정조(DEX조,제전30 min주사DEX),유생소C조(VC조,제전10 min주사VC),우미탁미정연합유생소C조( DEX+VC조,제전30 min주사DEX,제전10 min주사VC)。결혈20 min재관주24 h후취소장조직,소목정-이홍염색관찰소장점막적손상정황,면역조화법검측소장점막중백세포개소-6(IL-6)、종류배사인자α(TNF-α)화caspase-3적표체변화병주chiu′s평분。결과 DEX、VC、DEX+VC조소장점막적chiu′s평분화IL-6、TNF-α、caspase-3적표체교I/R조강저(P<0.05),이교sham조승고(P<0.05);DEX조화VC조차이무통계학의의( P>0.05),DEX +VC조소장점막적chiu′s평분화IL-6、TNF-α、caspase-3적표체교DEX、VC조강저(P<0.05)。결론우미탁미정、유생소C대소서소장결혈재관주손상유보호작용,량자련용유협동작용,차보호작용여IL-6,TNF-α화caspase-3유관。
Objective To investigate the protective effects of dexmedetomidine ( DEX) , vitamin C ( VC) or both a-gainst intestinal ischemia/reperfusion in mice and possible mechanism .Methods A total of 50 mice were randomly di-vided into five groups (n=10) according to their treatments:a sham group (in which the superior mesenteric artery was exposed without ligation ), an intestinal ischemia/reperfusion ( I/R) group ( which underwent ischemia for 20 min fol-lowed by reperfusion for 24 h), a dexmedetomidine (DEX) group (which received DEX 30 min before intestinal I/R, followed by ischemia for 20 min and consequently reperfusion for 24 h) , a vitamin C ( VC) group ( which received vita-min C 10 min before intestinal I/R, followed by ischemia for 20 min and consequently reperfusion for 24 h) , and a com-bined treatment ( DEX+VC) group ( which were injected with DEX and VC 30 min and 10 min before intestinal I/R re-spectively, followed by ischemia for 20 min and consequently reperfusion for 24 h).The pathological changes of the small intestinal mucosa were observed by HE staining .The amounts of IL-6, TNF-αand caspase-3 were detected by im-munohistochemistry and analyzed by Chiu′s scoring assessment .Results Groups DEX, VC, and DEX+VC produced the scores of Chiu′s ratings and the amounts of IL -6, TNF-αand caspase-3 which were lower than Group I/R (P<0.05), but higher than Group sham (P<0.05).But no statistical difference was determined between Groups DEX and VC (P>0.05).Group DEX+VC produced reduced Chiu′s scores and the amounts of IL -6, TNF-αand caspase-3 when compared with Groups DEX and VC (P<0.05).Conclusion Dexmedetomidine and vitamin C attenuate the in-testinal ischemia/reperfusion injury in mice , which can result in synergism related with IL -6, TNF-αand caspase-3.
