采集拙政园3个底泥和3个土壤样品,运用末端限制性片段长度多态性(T-RFLP)技术分析拙政园6个样品的微生物多样性。研究结果显示:底泥细菌中T-RFs片段大小为181 bp、211 bp的菌种为优势菌种,土壤细菌中T-RFs片段大小为91 bp、205 bp的菌种为优势菌种。各样点底泥的真菌种类各不相同,且种类较少,无明显的优势菌种,而土壤真菌中T-RFs片段大小为355 bp的菌种为优势菌种。这说明拙政园底泥、土壤的细菌和真菌多样性存在差异,细菌多样性明显高于真菌。为苏州园林微生物多样性及群落结构累积了有价值的资料。

Terminal restriction fragment length polymorphism (T-RFLP) technique was used to analyze microbial diversity in 3 points of sediment and 3 points of soil in Humble Administrator's Garden. The results show that a-mong the sediment bacteria ,T-RFs fragments with the sizes of 181 bp ,211 bp were the dominant bacteria. A-mong soil bacteria ,T-RFs fragments with the sizes of 91 bp ,205 bp were the dominant bacteria. Among sediment fungi, species of each site were not identical. Among soil fungi ,T-RFs fragment with the size of 355 bp was the dominant fungus. Bacterial and fungal diversity of sediment and soil in Humble Administrator's Garden were dif-ferent. Bacterial diversity was significantly higher than that of fungi. This study has provided valuable data for microbial diversity and community structure of Suzhou gardens.
