以新鲜石榴皮为原料,对水提醇沉法提取的石榴皮粗多糖进行三氯乙酸法脱蛋白工艺研究。对三氯乙酸浓度、振摇时间以及除蛋白次数等影响脱蛋白效率的因素,采取单因素和L9(33)正交试验进行研究。结果表明,三氯乙酸法脱除石榴皮多糖蛋白质的最佳工艺条件为:三氯乙酸浓度5%,振摇时间10 min,脱蛋白次数3次,此条件下石榴皮多糖中蛋白率脱除率达89.0%,多糖损失率为8.4%,可作为是一种有效的脱除石榴皮粗多糖蛋白质的方法。

Pomegranate peel polysaccharide was obtained from pomegranate peel by water-alcohol extraction. Trichloroacetic acid method was used to take off the protein in the crude polysaccharides from pomegranate peel. The four parameters including the concentration of trichloroacetic acid, the shaking time and the number of times for deproteinization were studied. The single factor experiment and a L9(33) orthogonal experiment including these factors were carried out to determine the optimal condition of deproteinization. The results showed that the optimum conditions of deproteinization were obtained as follows:the concentration of 5% trichloroacetic acid added, the shaking time of 10 min, and the deproteinization times of three. Under these conditions, the rate of deproteinization can get to 89. 0% and the loss rate of polysaccharide reach 8. 4%. It can be taken as an effective deproteinization method for crude polysaccharide from pomegranate peel.
