为获得高产杆菌肽菌株,对海水养殖环境中分离的产杆菌肽菌株地衣芽孢杆菌ZZ061进行A T RP诱变,以藤黄微球菌为指示菌,采用抑菌圈法进行筛选,获得一株产杆菌肽水平明显提高的诱变株S F1406,其摇瓶效价比野生菌株提高130%,达到560U·mL -1。采用正交试验优化S F1406产杆菌肽培养基,结果表明:在玉米淀粉含量为3%,硫酸铵含量为1.5%,豆饼粉含量为1%,蔗糖含量为3%时效果最佳,杆菌肽效价进一步提高44%,达到810U·mL -1。
為穫得高產桿菌肽菌株,對海水養殖環境中分離的產桿菌肽菌株地衣芽孢桿菌ZZ061進行A T RP誘變,以籐黃微毬菌為指示菌,採用抑菌圈法進行篩選,穫得一株產桿菌肽水平明顯提高的誘變株S F1406,其搖瓶效價比野生菌株提高130%,達到560U·mL -1。採用正交試驗優化S F1406產桿菌肽培養基,結果錶明:在玉米澱粉含量為3%,硫痠銨含量為1.5%,豆餅粉含量為1%,蔗糖含量為3%時效果最佳,桿菌肽效價進一步提高44%,達到810U·mL -1。

The bacitracin‐producing Bacillus licheniformis ZZ061 was isolated from an aquaculture facility .From the isolated bacteria , mutants with enhanced capability of generating bacitrcin were bred by means of ARTP mutagenesis .In the process ,Micrococcus luteus was used as the indicator bacterium ,and the bacteriostatic circle method employed for the screening .One of the mutants ,SF1406 ,showed the greatest increase ,i .e .,130% higher than the wild strain , in its bacitrcin productivity . Subsequently , an orthogonal experiment was conducted to optimize the culture medium formulation .It was found that the medium consisting of 3% corn starch ,1.5%ammonium sulfate ,1% soybean powder and 3% sucrose rendered the most desirable results with an additional 44%productivity increase on the yield of 810 U · mL -1 .
