目的:探讨抗抑郁剂盐酸帕罗西汀对卵巢切除大鼠学习记忆能力、血清雌激素水平及大鼠海马雌激素受体表达的影响。方法采用 SD 雌鼠双侧卵巢切除(OVX)作为雌激素波动引起的抑郁与认知障碍模型,用 Morris 水迷宫实验来测试大鼠的学习记忆功能。实验共分为正常对照组、OVX对照组、OVX 药物组。予5-羟色胺重摄取抑制剂类药物:盐酸帕罗西汀10 mg/(kg·d)干预4周。采用 ELISA 法测量SD 大鼠血清黄体生成素、卵泡刺激素、雌激素。免疫组化法检测大鼠海马雌激素受体α(ERα)和雌激素受体β(ERβ)的阳性细胞表达数。结果① Morris 水迷宫实验中,OVX对照组和 OVX 药物组的逃避潜伏期明显缩短,停留在平台时间百分比明显增加。② OVX 药物组的血清雌激素较OVX 对照组明显增加(P <0.05)。③药物组与非药物组的ERα、ERβ表达无明显差异。结论帕罗西汀增加OVX 大鼠血清雌激素水平和改善学习记忆能力,而对海马ERα、ERβ表达无明显影响。

목적:탐토항억욱제염산파라서정대란소절제대서학습기억능력、혈청자격소수평급대서해마자격소수체표체적영향。방법채용 SD 자서쌍측란소절제(OVX)작위자격소파동인기적억욱여인지장애모형,용 Morris 수미궁실험래측시대서적학습기억공능。실험공분위정상대조조、OVX대조조、OVX 약물조。여5-간색알중섭취억제제류약물:염산파라서정10 mg/(kg·d)간예4주。채용 ELISA 법측량SD 대서혈청황체생성소、란포자격소、자격소。면역조화법검측대서해마자격소수체α(ERα)화자격소수체β(ERβ)적양성세포표체수。결과① Morris 수미궁실험중,OVX대조조화 OVX 약물조적도피잠복기명현축단,정류재평태시간백분비명현증가。② OVX 약물조적혈청자격소교OVX 대조조명현증가(P <0.05)。③약물조여비약물조적ERα、ERβ표체무명현차이。결론파라서정증가OVX 대서혈청자격소수평화개선학습기억능력,이대해마ERα、ERβ표체무명현영향。
Objective To observe whether paroxetine affects rats’learning and memory ability,serum estrogen and the expression of estrogen receptors in hippocampus.Methods The SD ovariectomized(OVX)rats were a-dopted as the model of estrogen's fluctuation leading to depressive symptoms and cognition disorder,treated with paroxetine by intragastric administration at 10 mg/(kg·d)for 4 weeks,and the sham rats as control group.Morris water maze was used to testify the effect of paroxetine on learning and memory ability.The level of serum luteinizing hormone,follicle-stimulating hormone,estrogen was measured by ELISA.Expressions of ERαand ERβin hipp-ocampal neurons were detected by immunohistochemistry.Results ① In Morris water maze experiment,compared to the control group,the escape latency of two drug-groups were obviously shortened;the time of staying in the plat-form significantly increased.② Serum estrogen of drug-group was significantly increased compared with the control groups (P <0.05).③ The ER expression between groups treated with paroxetine and the group without paroxetine in OVX rats had no difference.Conclusion Paroxetine increses serum estrogen levels and improves the learning and memory ability,and has no significant effect on ER expression in hippocampal neurons.
