目的:观察并分析1,25-二羟维生素 D3对免疫性血小板减少症(ITP)模型小鼠的治疗作用。方法将44只BALB/c 小鼠分为正常对照组、模型组和1,25-二羟维生素D3治疗组。模型组经腹腔给予豚鼠抗小鼠血小板血清(GP-APS),治疗组在模型组的基础上经尾静脉注射给予100 ng/d 的1,25-二羟维生素 D3,正常对照组均给予相同体积的生理盐水经腹腔及尾静脉注射给药。在每次给药后24 h 记录体重并采血,测量血小板计数,评价出血倾向。2周后处死小鼠并解剖,取骨髓涂片,镜下计数巨核细胞。从多方面评估1,25-二羟维生素 D3对模型小鼠的治疗作用。结果注射第10天开始,1,25-二羟维生素 D3治疗组小鼠进食量开始明显增加,精神和反应逐渐恢复;体重、血小板数较模型组明显上升(P <0.05);出血倾向较模型组明显降低(P <0.05);产板巨核细胞数较模型组高,低于正常对照组,而巨核细胞数较模型组低,高于正常对照组,差异有统计学意义。结论1,25-二羟维生素D3对模型小鼠在一般情况的改善、体重及血小板数的上升、出血倾向的降低和促进骨髓血小板生成等方面表现出一定影响。

목적:관찰병분석1,25-이간유생소 D3대면역성혈소판감소증(ITP)모형소서적치료작용。방법장44지BALB/c 소서분위정상대조조、모형조화1,25-이간유생소D3치료조。모형조경복강급여돈서항소서혈소판혈청(GP-APS),치료조재모형조적기출상경미정맥주사급여100 ng/d 적1,25-이간유생소 D3,정상대조조균급여상동체적적생리염수경복강급미정맥주사급약。재매차급약후24 h 기록체중병채혈,측량혈소판계수,평개출혈경향。2주후처사소서병해부,취골수도편,경하계수거핵세포。종다방면평고1,25-이간유생소 D3대모형소서적치료작용。결과주사제10천개시,1,25-이간유생소 D3치료조소서진식량개시명현증가,정신화반응축점회복;체중、혈소판수교모형조명현상승(P <0.05);출혈경향교모형조명현강저(P <0.05);산판거핵세포수교모형조고,저우정상대조조,이거핵세포수교모형조저,고우정상대조조,차이유통계학의의。결론1,25-이간유생소D3대모형소서재일반정황적개선、체중급혈소판수적상승、출혈경향적강저화촉진골수혈소판생성등방면표현출일정영향。
Objective To observe and analyze the therapeutic effect of 1,25-dihydroxyvitamin D3 on the immune thrombocytopenia(ITP)model in mice.Methods 44 BALB /c mice were divided into three groups:normal control group,model group and 1,25-dihydroxyvitamin D3 treatment group.The model group was given the GP-APS intra-peritoneally,while the treatment group,on the basis of the model group,was administered 100 ng/d of 1,25-di-hydroxyvitamin D3 by intravenous injection,and the normal control group was given the same volume of saline through both intraperitoneal and intravenous injection.The mice were taken blood samples after 24 hours of each administration,recorded their weights and measured platelet accounts and bleeding tendencies.After two weeks of injection,the mice were sacrificed and dissected,smeared with the bone marrow that had been taken out,counted the number of megakaryocyte under the microscope.Assessed the therapeutic effect of 1,25-dihydroxyvitamin D3 on the model mice in several aspects.Results From the 10th day of the injection,the food-intake of the treatment group was markedly increased,the spirit status and reaction was gradually recovered;the body weight and platelet account,compared with the model group,were both significantly increased (P <0.05);bleeding tendency evident was markedly decreased (P <0.05);the number of the platelet-forming megakaryocyte was higher than the model group and lower than the normal control group,while the number of the megakaryocyte was lower than the model group and higher than the normal control group,the difference was statistically significant.Conclusion 1,25-di-hydroxyvitamin D3 shows some influence on the improvement of general status,the rise of body weight and platelet account,the reduction of bleeding tendency and the promotion of platelet's production of the model mice.
