目的：分析日本血吸虫感染以及虫源性糖蛋白果糖二磷酸醛缩酶（SjFBPA）刺激后小鼠脾脏细胞中白介素4（IL-4）、白细胞分化抗原 CD8a 因子表达量的动态变化，并探讨 SjFBPA 与辅助型 T 细胞（Th2）免疫应答的关系。方法血吸虫尾蚴感染 C57小鼠10只，设正常对照组。感染8周后处死小鼠，取脾脏制备淋巴细胞悬液；用藻红蛋白（PE）标记 CD8a 抗体，藻青蛋白（APC）标记 IL-4抗体进行细胞染色；流式细胞仪检测细胞因子表达量的变化。10只正常 C57小鼠处死后取脾脏制成淋巴细胞悬液，加 SjFBPA 蛋白体外刺激，按上述流程刺激方法检测细胞。结果小鼠感染血吸虫8周后脾细胞中 IL-4、CD8a 的表达量分别为（22．39±5．93）％、（11．34±2．15）％，正常对照组脾细胞中 IL-4、CD8a 的表达量分别为（1．39±0．52）％、（1．98±0．84）％，差异有统计学意义（P ＜0．01）。SjFBPA 体外刺激小鼠脾细胞的 IL-4、CD8a 的表达量分别为（14．14±1．16）％、（8．99±0．81）％，正常对照组脾细胞中 IL-4、CD8a 的表达量分别为（1．86±0．93）％、（1．69±0．56）％，差异有统计学意义（P ＜0．01）。结论 SjFBPA 在血吸虫感染中具有诱导 Th2免疫偏移的作用，树突状细胞作为提呈细胞参与该免疫反应。
목적：분석일본혈흡충감염이급충원성당단백과당이린산철축매（SjFBPA）자격후소서비장세포중백개소4（IL-4）、백세포분화항원 CD8a 인자표체량적동태변화，병탐토 SjFBPA 여보조형 T 세포（Th2）면역응답적관계。방법혈흡충미유감염 C57소서10지，설정상대조조。감염8주후처사소서，취비장제비림파세포현액；용조홍단백（PE）표기 CD8a 항체，조청단백（APC）표기 IL-4항체진행세포염색；류식세포의검측세포인자표체량적변화。10지정상 C57소서처사후취비장제성림파세포현액，가 SjFBPA 단백체외자격，안상술류정자격방법검측세포。결과소서감염혈흡충8주후비세포중 IL-4、CD8a 적표체량분별위（22．39±5．93）％、（11．34±2．15）％，정상대조조비세포중 IL-4、CD8a 적표체량분별위（1．39±0．52）％、（1．98±0．84）％，차이유통계학의의（P ＜0．01）。SjFBPA 체외자격소서비세포적 IL-4、CD8a 적표체량분별위（14．14±1．16）％、（8．99±0．81）％，정상대조조비세포중 IL-4、CD8a 적표체량분별위（1．86±0．93）％、（1．69±0．56）％，차이유통계학의의（P ＜0．01）。결론 SjFBPA 재혈흡충감염중구유유도 Th2면역편이적작용，수돌상세포작위제정세포삼여해면역반응。
Objective To analyze the dynamics of IL-4,CD8a in spleen cells of mice infected with cercariae of Schistosoma japonicum or SjFBPA and investigate the relationship between SjFBPA and the Th2-dominant response. Methods 10 mice were infected by cercariae and 10 mice were uninfected.8 weeks after infection,all mice were sacrificed.Spleen single cell suspension was prepared and analyzed by flowed cytometry.Analysis was performed on the IL-4,CD8a cells of spleen cells stained with anti FITC-CD4,PE-CD8a,APC-IL-4 monoclonal antibody.Re-sults At 8 weeks,the percentage of IL-4 and CD8a cells in infected with cercariae of Schistosoma japonicum mice was 24.5% and 7.65%,respectively,while the percentage of IL-4 and CD8a cells in uninfected mice was 1.18%and 0.571%,indicating a gradual decline (P <0.05).The percentage of IL-4 and CD8a cells in infected with SjFBPA was 18.3% and 5.82%,the percentage of IL-4 and CD8a cells in uninfected mice was 0.91% and 0.086%,indicating a gradual decline (P <0.05).Conclusion This study suggests that SjFBPA may have a po-tential value for the Th2 response.DC cells as presenting cells are involved in the immune response.
