目的 分析EB病毒( EBV) miRNAs在鼻咽癌组织中的表达,探索EBV诱发鼻咽癌的分子机制,寻找鼻咽癌防治和诊断的生物分子标志物. 方法 采用实时荧光定量PCR分析 EBV miRNAs ebv-miR-BART4?和 ebv-miR-BART18-3p在36例鼻咽低分化鳞癌组织和32例慢性鼻咽黏膜炎症组织中的表达水平. 结果 ebv-miR-BART4? 和 ebv-miR-BART18-3p的表达水平( Ct 值)在鼻咽癌组织中分别为(26. 97 ± 2. 67)和(27. 02 ± 2. 32),在鼻咽黏膜炎症组织中的表达水平分别为( 38. 49 ± 3. 05 )和( 39. 39 ± 2. 16 ) ,两种EBV miRNAs在鼻咽癌组织中的表达明显高于鼻咽黏膜炎症组织,差异有统计学意义( P<0. 01 ). 结论 EBV可能通过ebv-miR-BART4?和ebv-miR-BART18-3p高表达诱发鼻咽癌,它们是鼻咽癌诊断和防治的潜在生物分子标志物.

Objective To analyze expression of EB virus ( EBV) miRNAs in nasopharyngeal carcinoma( NPC) tis-sue, explore molecular mechanism of EBV induced NPC and biomarker for prevention and diagnosis of NPC. Meth-ods 36 cases of nasopharyngeal poorly differentiated squamous cell carcinoma tissues and 32 cases of chronic na-sopharyngeal mucositis tissues were collected. Expressions of EBV miRNAs ebv-miR-BART4? and ebv-miR-BART18-3 p were tested in these tissues by quantitative real-time PCR; the expression level of EBV miRNAs was analyzed in both tissue groups. Results The expressions( Ct) of ebv-miR-BART4? and ebv-miR-BART18-3p were (26. 97 ± 2. 67) and (27. 02 ± 2. 32) in NPC tissue, (38. 49 ± 3. 05) and (39. 39 ± 2. 16)in nasopharyngeal mu-cosa inflammation tissue, the expressions of two ebv-miRNAs were significantly higher in NPC tissue than that in nasopharyngeal mucosa inflammation tissue ( P <0. 01 ) . Conclusion NPC may be induced by EBV through the high expression of ebv-miR-BART4?and ebv-miR-BART18-3p. They are potential biological molecular markers for the diagnosis and treatment of NPC.
