目的:观察补肾中药对肾虚质大鼠脑源性神经营养因子(BDNF)表达的影响,探讨生物体海马区神经元功能活动与肾藏志理论相关性。方法采用“猫吓鼠”经典造模法造成先天不足加后天失养复合型肾虚质模型仔鼠,分为模型组、左归丸组、右归丸组,空白组产自正常孕鼠,选用补肾中药进行调理。检测大鼠脑质量、肾质量、脑重指数及海马区BDNF的表达。结果与空白组比较,肾虚质大鼠脑质量、肾质量、脑重指数和BDNF光密度值降低;镜下观察海马区出现片状水肿,神经细胞数目减少、结构松散。补肾中药干预后,脑质量、肾质量、脑重指数及BDNF光密度值得到一定程度回升;大脑皮质包括海马区神经细胞增多,细胞结构致密且层次清楚,无明显水肿区。结论大鼠海马 BDNF 表达变化与肾藏志理论具有相关性;补肾中药可显著改善肾虚质大鼠脑功能衰退状况。
目的:觀察補腎中藥對腎虛質大鼠腦源性神經營養因子(BDNF)錶達的影響,探討生物體海馬區神經元功能活動與腎藏誌理論相關性。方法採用“貓嚇鼠”經典造模法造成先天不足加後天失養複閤型腎虛質模型仔鼠,分為模型組、左歸汍組、右歸汍組,空白組產自正常孕鼠,選用補腎中藥進行調理。檢測大鼠腦質量、腎質量、腦重指數及海馬區BDNF的錶達。結果與空白組比較,腎虛質大鼠腦質量、腎質量、腦重指數和BDNF光密度值降低;鏡下觀察海馬區齣現片狀水腫,神經細胞數目減少、結構鬆散。補腎中藥榦預後,腦質量、腎質量、腦重指數及BDNF光密度值得到一定程度迴升;大腦皮質包括海馬區神經細胞增多,細胞結構緻密且層次清楚,無明顯水腫區。結論大鼠海馬 BDNF 錶達變化與腎藏誌理論具有相關性;補腎中藥可顯著改善腎虛質大鼠腦功能衰退狀況。
목적:관찰보신중약대신허질대서뇌원성신경영양인자(BDNF)표체적영향,탐토생물체해마구신경원공능활동여신장지이론상관성。방법채용“묘하서”경전조모법조성선천불족가후천실양복합형신허질모형자서,분위모형조、좌귀환조、우귀환조,공백조산자정상잉서,선용보신중약진행조리。검측대서뇌질량、신질량、뇌중지수급해마구BDNF적표체。결과여공백조비교,신허질대서뇌질량、신질량、뇌중지수화BDNF광밀도치강저;경하관찰해마구출현편상수종,신경세포수목감소、결구송산。보신중약간예후,뇌질량、신질량、뇌중지수급BDNF광밀도치득도일정정도회승;대뇌피질포괄해마구신경세포증다,세포결구치밀차층차청초,무명현수종구。결론대서해마 BDNF 표체변화여신장지이론구유상관성;보신중약가현저개선신허질대서뇌공능쇠퇴상황。
Objective To observe the effects of kidney-supplementing TCM on the brain-derived neurotrophic factor (BDNF) expression of rats with kidney-deficiency constitution;To discuss the correlation between functional activity of neurons in hippocampus with the theory of kidney storing aspiration.MethodsComposite rat models with offspring deficiency and acquired dystrophy were built through the method of “cats scare rats”, and then were divided into model group,Zuogui Pill group,Yougui Pill group, and blank group in which offspring came from normal pregnant rats. After the treatment with kidney-supplementing TCM, weight of brain and kidney, brain weight indexes and the BDNF expression were detected.Results Compared with the blank group, the quality of brain, kidney and brain weight indexes, BDNF expression decteased of rats in model group were lower;flaky edema appeared in hippocampus observed by microscope;the number of nerve cells decreased and loosely structured. After the treatment of kidney-supplementing TCM, the weight of brain and kidney and brain weight indexes recovered to some extent;BDNF expression increased;nerve cells in cerebral cortexincluding hippocampus increased;the cell structure compacted and the level cleared as well as no edema.Conclusion The BDNF expression has correlation with the theory of kidney storing aspiration. Kidney-supplementing TCM can improve brain function and significantly improve declining situation of brain function of rats with kidney-deficiency constitution.
