目的 研究凝血酶敏感激素蛋白酶4/5 ( ADAMTS4/5)在骨性关节炎( OA)滑膜组织及滑膜细胞中的表达及意义. 方法 取6例正常者滑膜组织和6 例OA患者滑膜组织,Western blot法检测标本中ADAMTS4/5 的表达,离体培养正常者滑膜细胞. 白介素-1β( IL-1β)刺激正常滑膜细胞,测定滑膜细胞中ADAMTS4/5的表达量;IL-1RA( IL-1β阻滞剂)抑制IL-1β后,测定滑膜细胞中ADAMTS4/5的表达量.结果 ADAMTS4/5 在OA患者滑膜组织中表达较正常者滑膜组织均显著增加;IL-1β刺激滑膜细胞后,ADAMTS4/5升高,IL-1RA可明显降低IL-1β诱导的滑膜细胞中ADAMTS4/5的表达. 结论 IL-1β可以按照剂量依赖方式正向调节ADAMTS4/5的表达;ADAMTS4/5 和IL-1β在OA病变机制中发挥重要的作用.

목적 연구응혈매민감격소단백매4/5 ( ADAMTS4/5)재골성관절염( OA)활막조직급활막세포중적표체급의의. 방법 취6례정상자활막조직화6 례OA환자활막조직,Western blot법검측표본중ADAMTS4/5 적표체,리체배양정상자활막세포. 백개소-1β( IL-1β)자격정상활막세포,측정활막세포중ADAMTS4/5적표체량;IL-1RA( IL-1β조체제)억제IL-1β후,측정활막세포중ADAMTS4/5적표체량.결과 ADAMTS4/5 재OA환자활막조직중표체교정상자활막조직균현저증가;IL-1β자격활막세포후,ADAMTS4/5승고,IL-1RA가명현강저IL-1β유도적활막세포중ADAMTS4/5적표체. 결론 IL-1β가이안조제량의뢰방식정향조절ADAMTS4/5적표체;ADAMTS4/5 화IL-1β재OA병변궤제중발휘중요적작용.
Objective To research the expression of thrombin sensitive hormone protease 4 ( ADAMTS4/5 ) in the OA synovial tissue and cells. Methods 6 cases of OA synovial tissue and 6 cases of normal synovial tissue were taken during the operation. The expression quantity of specimen ADAMTS4/5 in cases was measured by Western blot, respectively. The normal group and OA group synovial cells were cultured in vitro. The normal group synovial cells were induced by IL-1βor IL-1βblockers (IL-1RA). The expression quantity of ADAMTS4/5 were measured by the supernatant fluid. Results ADAMTS4/5 in OA synovial tissue expressed significantly than the normal group. The group of IL-1β was obviously higher, IL-1RA could block IL-1β and induce synovial cells to produce ADAMTS4/5 . Conclusion ADAMTS4/5 and IL-1βplay an important role in synovial tissue inflammatory change;IL-1β can be in accordance with the dose-response manner positive adjustment ADAMTS4/5 expression.
