采用微波结合链霉素抗性筛选法选育放线菌素 D 的高产菌株。通过考察链霉素对 Streptomyces rubiginosohelvolus FIM-N31菌株孢子生长情况的影响确定链霉素致死浓度,出发菌株FIM-N31的孢子经微波辐照处理后,涂布在含链霉素致死浓度(50μg/mL)的培养基平板上培养,获得了大量的链霉素抗性基因突变株。摇瓶发酵筛选突变株,结果获得一株遗传性状稳定的放线菌素D高产菌Str186,其产放线菌素D的能力比出发菌株提高了8倍以上。
採用微波結閤鏈黴素抗性篩選法選育放線菌素 D 的高產菌株。通過攷察鏈黴素對 Streptomyces rubiginosohelvolus FIM-N31菌株孢子生長情況的影響確定鏈黴素緻死濃度,齣髮菌株FIM-N31的孢子經微波輻照處理後,塗佈在含鏈黴素緻死濃度(50μg/mL)的培養基平闆上培養,穫得瞭大量的鏈黴素抗性基因突變株。搖瓶髮酵篩選突變株,結果穫得一株遺傳性狀穩定的放線菌素D高產菌Str186,其產放線菌素D的能力比齣髮菌株提高瞭8倍以上。
채용미파결합련매소항성사선법선육방선균소 D 적고산균주。통과고찰련매소대 Streptomyces rubiginosohelvolus FIM-N31균주포자생장정황적영향학정련매소치사농도,출발균주FIM-N31적포자경미파복조처리후,도포재함련매소치사농도(50μg/mL)적배양기평판상배양,획득료대량적련매소항성기인돌변주。요병발효사선돌변주,결과획득일주유전성상은정적방선균소D고산균Str186,기산방선균소D적능력비출발균주제고료8배이상。
Microwave mutagenesis and streptomycin resistance screening was applied to screen actinomycin D high-producting strain.After testing the resistance of Streptomyce rubiginosohelvolus FIM-N31 to streptomycin, the lethal concentration of streptomycin was determined.A lot of streptomycin-resistant mutants were screened after the spores regenerated on the lethal streptomycin concentration ( 50μg/mL) media when they were treated with microwave irradiation.It was eventually found that the high-yield strain Str186 from these mutants which was stably inheritable, and the productivity was beyond eight times higher than the original strains in the rotation-flask experiments.
