目的 观察父源性( F0 代雄性小鼠)十溴联苯醚(PBDE-209)暴露对胎鼠(F1 代)体格发育状况及出生后70 (PND70)天F1 代雄性子鼠脏器系数、睾酮水平、精子数和睾丸组织形态的影响. 方法 4周龄雄性小鼠,按小鼠每日体重进行灌胃染毒,随机分为玉米油组( 0 mg/kg )、低剂量组(100 mg/kg)、中剂量组(300 mg/kg)和高剂量组(500 mg/kg) ,连续染毒6周. 染毒结束前1 d与健康雌鼠1: 2合笼,观察F1 代胎鼠体格发育和PND70天F1 代雄性子鼠脏器系数、睾酮水平、精子数和睾丸组织形态变化. 结果 高剂量组F1 代胎鼠体重、体长和肛殖距(肛门到生殖器距离)降低(P<0. 05),雌雄比增加(P<0. 05);与玉米油组比较,高剂量组PND70天雄性子鼠睾丸系数、附睾系数、肝脏系数、肾脏系数、附睾尾精子数和血清睾酮水平明显降低(P <0. 05);雄性仔鼠HE染色显示中、高剂量组精子数目减少,间质间隙明显增宽,基底室出现空腔,支持细胞和间质细胞数减少等. 结论 F0 代雄性小鼠暴露PBDE-209 可能会影响F1 代子鼠体格发育,还可能会使PND70天雄性子鼠的精子数、睾酮水平降低,以及睾丸形态结构发生改变.

목적 관찰부원성( F0 대웅성소서)십추련분미(PBDE-209)폭로대태서(F1 대)체격발육상황급출생후70 (PND70)천F1 대웅성자서장기계수、고동수평、정자수화고환조직형태적영향. 방법 4주령웅성소서,안소서매일체중진행관위염독,수궤분위옥미유조( 0 mg/kg )、저제량조(100 mg/kg)、중제량조(300 mg/kg)화고제량조(500 mg/kg) ,련속염독6주. 염독결속전1 d여건강자서1: 2합롱,관찰F1 대태서체격발육화PND70천F1 대웅성자서장기계수、고동수평、정자수화고환조직형태변화. 결과 고제량조F1 대태서체중、체장화항식거(항문도생식기거리)강저(P<0. 05),자웅비증가(P<0. 05);여옥미유조비교,고제량조PND70천웅성자서고환계수、부고계수、간장계수、신장계수、부고미정자수화혈청고동수평명현강저(P <0. 05);웅성자서HE염색현시중、고제량조정자수목감소,간질간극명현증관,기저실출현공강,지지세포화간질세포수감소등. 결론 F0 대웅성소서폭로PBDE-209 가능회영향F1 대자서체격발육,환가능회사PND70천웅성자서적정자수、고동수평강저,이급고환형태결구발생개변.
Objective To observe the physical development of fetal mice ( F1 fetal mice) and the organ coefficient, testosterone levels, sperm count and testicular tissue morphology of F1 male offspring after birth 70 (PND70) days by the paternal (F0 male mice) exposing to decabromodiphenyl ether (PBDE-209). Methods 4-week-old male mice were infected according to the daily body weight and divided into the corn oil group ( 0 mg/kg ) , low dose group (100 mg/kg), middle dose group (300 mg/kg) and high dose group (500 mg/kg) randomly. Mice were exposed to PBDE-209 by gavage for 6 weeks. They were put together with healthy female mice for copulation as the ratio 1: 2 before the end of gavage. To observe the physical development of F1 fetal mice and the organ coefficient, testosterone levels, sperm count and testicular tissue morphology of F1 male offspring after birth 70 (PND70) days. Results In the high dose group, the body weight, length and anogenital distance ( distance between the anus to the genitals) of F1 fetal mice were reduced (P<0. 05), and the ratio of female verse male increased (P<0. 05);compared with the control group, the testis coefficient, epididymis coefficient, liver coefficient and kidney coeffi-cient of the high-dose group PND70 male offspring were decreased ( P<0. 05 ) , and the sperm count in the tail of epididymal and serum testosterone levels were reduced significantly ( P <0. 05 ); in the middle and high dose groups, the number of sperm count was reduced, the interstitial space was widened significantly, vacuolus appeared in the base compartment, and the number of sertoli cells and leydig cells was reduced in the male fetal mice by HE staining. Conclusion F0 male mice exposed to PBDE-209 may affect the physical development of F1 fetal mice, and may cause the reduction of sperm count and testosterone levels and testicular morphological change of PND70 male offspring.
