为研究促炎性细胞因子 IL-17是否参与羊口疮炎症的发生,用羊口疮强、弱毒株分别体外刺激山羊外周血单个核淋巴细胞(PBMCs),采用 real-time PCR 方法检测 IL-17及与 Th17细胞分化并活化相关的细胞因子 mRNA 水平。结果表明用羊口疮病毒强毒株刺激山羊 PBMCs 后,IL-17转录水平升高;与Th17细胞分化并活化相关的细胞因子如 TGF-β、IL-1β、IL-6和 IL-23P19转录水平变化明显。证明羊口疮病毒强毒株能刺激羊 PBMCs 增殖,使初始 T 细胞分化为 Th17细胞并活化分泌 IL-17,从而介导炎症反应。
為研究促炎性細胞因子 IL-17是否參與羊口瘡炎癥的髮生,用羊口瘡彊、弱毒株分彆體外刺激山羊外週血單箇覈淋巴細胞(PBMCs),採用 real-time PCR 方法檢測 IL-17及與 Th17細胞分化併活化相關的細胞因子 mRNA 水平。結果錶明用羊口瘡病毒彊毒株刺激山羊 PBMCs 後,IL-17轉錄水平升高;與Th17細胞分化併活化相關的細胞因子如 TGF-β、IL-1β、IL-6和 IL-23P19轉錄水平變化明顯。證明羊口瘡病毒彊毒株能刺激羊 PBMCs 增殖,使初始 T 細胞分化為 Th17細胞併活化分泌 IL-17,從而介導炎癥反應。
위연구촉염성세포인자 IL-17시부삼여양구창염증적발생,용양구창강、약독주분별체외자격산양외주혈단개핵림파세포(PBMCs),채용 real-time PCR 방법검측 IL-17급여 Th17세포분화병활화상관적세포인자 mRNA 수평。결과표명용양구창병독강독주자격산양 PBMCs 후,IL-17전록수평승고;여Th17세포분화병활화상관적세포인자여 TGF-β、IL-1β、IL-6화 IL-23P19전록수평변화명현。증명양구창병독강독주능자격양 PBMCs 증식,사초시 T 세포분화위 Th17세포병활화분비 IL-17,종이개도염증반응。
For investigating weather the pro-inflammatory cytokine IL-17 participates inflammation occur-rence of ORF,the virulent ORF virus and attenuated ORF virus were used to stimulate the peripheral blood mononuclear cells in goats,respectively.Then transcription levels of IL-17 and cytokines were detected by real-time PCR method.The result showed that the transcription level of IL-17 increased after the lympho-cytes being stimulated by virulent ORF virus.In addition,the transcription levels of cytokines,relating with differentiation and activation of Th17 cell,TGF-β,IL-1β,IL-6 and IL-23P19 changed obviously.This re-search indicated that ORF virus stimulated lymphocyte proliferation,and Th17 cell differentiation from na-ive T cells,activated and secreted cytokine IL-17,and mediated inflammation occurrence.
