目的:本研究旨在观察高尿酸血症对小鼠脑缺血再灌注损伤的影响,了解高尿酸血症是否加重其损伤。方法取76只雄性昆明小鼠,随机分组为:A组(酵母膏+乙胺丁醇)、B组(酵母膏+乙胺丁醇+尼莫地平);C组(对照组)。A组采用酵母膏+乙胺丁醇,B组采用酵母膏+乙胺丁醇+尼莫地平,分别连续灌胃3周制作高尿酸血症模型小鼠,C组蒸馏水灌胃三周作为对照组。夹闭小鼠一侧颈总动脉2h后,再灌注22h制作小鼠脑缺血再灌注模型,并对各组小鼠:①按Longa法进行神经功能评分;②眶后静脉丛取血、测定血清尿酸、MDA、SOD水平,③断头取脑,观察脑组织形态学改变。结果实验结果显示:A组SOD活力显著降低,与C组(对照组)相比P=0.000,有显著性差异(P<0.01)。A组缺血脑组织内的MDA含量显著升高,与C组(对照组)相比为P=0.000,有显著性差异(P<0.01);脑组织形态学改变:A组可见神经细胞数明显减少,部分神经细胞只留其轮廓,总体表现为核固缩、碎裂、溶解等细胞坏死的特征。B组神经细胞损伤程度轻于A组。结论高尿酸血症加重脑缺血再灌注损伤。

목적:본연구지재관찰고뇨산혈증대소서뇌결혈재관주손상적영향,료해고뇨산혈증시부가중기손상。방법취76지웅성곤명소서,수궤분조위:A조(효모고+을알정순)、B조(효모고+을알정순+니막지평);C조(대조조)。A조채용효모고+을알정순,B조채용효모고+을알정순+니막지평,분별련속관위3주제작고뇨산혈증모형소서,C조증류수관위삼주작위대조조。협폐소서일측경총동맥2h후,재관주22h제작소서뇌결혈재관주모형,병대각조소서:①안Longa법진행신경공능평분;②광후정맥총취혈、측정혈청뇨산、MDA、SOD수평,③단두취뇌,관찰뇌조직형태학개변。결과실험결과현시:A조SOD활력현저강저,여C조(대조조)상비P=0.000,유현저성차이(P<0.01)。A조결혈뇌조직내적MDA함량현저승고,여C조(대조조)상비위P=0.000,유현저성차이(P<0.01);뇌조직형태학개변:A조가견신경세포수명현감소,부분신경세포지류기륜곽,총체표현위핵고축、쇄렬、용해등세포배사적특정。B조신경세포손상정도경우A조。결론고뇨산혈증가중뇌결혈재관주손상。
Objective To investigate the effects of Hyperuricemia on the rats with cerebral ischemia-reperfusion. MethodsThe healthy male Aba/c rats were randomly divided into three groups: A Group(treated with yeast extract and ethambutol intragastrically tree weeks)、B Group: (treated with yeast extract and ethambutol intragastrically tree weeks and Nimodipine、C Group: Control group ( given distilled water three weeks as a comparison) . After three weeks given nimodipine preoperative to group B, Ligate the carotid artery with slipknot to block the blood for 2 hours and then reperfusion for 22 hours to product the cerebral ischemia and reperfusion model by Longa method. Blood collection from Orbital venous plexus, decapitated the rats, detected the uric acid、SOD、MDA level of serum. Observe the morphological changes of the brain tissue.ResultsSOD levels significantly lower in serum of hyperuricemia model group, there ware significant difference compared with the control group (P<0.01).MDA levels were significantly elevated in the serum of model group,there were significant differences compared with the control group(P<0.01).ConclusionRats serum SOD、MDA level abnormalities may be associated with hyperuricemia aggravate cerebral ischemia-reperfusion injury.
