目的 探讨核因子-κB(NF-κB) p65在皮肤鳞状细胞癌及正常皮肤组织中的表达及意义.方法 采用反转录-聚合酶链反应(RT-PCR)和免疫组织化学链霉菌抗生物素蛋白-过氧化物酶(SP)法,检测40例鳞状细胞癌及40例正常皮肤组织中NF-κB p65 mRNA和蛋白的表达.结果 皮肤鳞状细胞癌组织中NF-κB p65 mRNA相对表达量(6.28±4.01)高于正常皮肤组织中的表达(1.62±0.99),组间比较差异有统计学意义(P<0.05);免疫组织化学结果显示皮肤鳞状细胞癌组织中NF-κB p65阳性表达率(58.7%)高于正常皮肤组织(11.8%),差异有统计学意义(x2=11.005,P<0.01).结论 NF-κB p65蛋白可能参与皮肤鳞状细胞癌的形成,减少NF-κB p65蛋白表达的靶向治疗.

Objective To determine whether nuclear transcription factor-κB (NF-κB) p65 expression was altered in skin squamous cell carcinoma (SCC) compared with normal skin.Methods We examined NF-κB p65 expression in 40 cases of SCC and compared to the normal skin (40 cases) by streptavid-in-peroxidase (SP) immunohistochemical methods and real-time quantitative polymerase chain reaction (Real-time PCR).Results The relative content of NF-κB p65 mRNA in SCC tissues (6.28 ± 4.01) was obviously higher than that (1.62 ±0.99) in normal skin tissues (P <0.05).The positive ratio of NF-κB p65 protein in SCC tissues was evidently higher than that in normal skin tissues (x2 =11.005,P < 0.01).Conclusion NF-κB p65 protein might be involved in formation of SCC.Target treatment of decreasing the expression of NF-κB p65 might be possible to prevent the formation of SCC.
