目的 分析肺腺癌原发病灶、癌旁组织、癌症脑转移病灶的基因表达.方法 收集1例患者肺腺癌原发病灶、癌旁组织、癌症脑转移病灶的组织样本进行测序,DEGseq算法筛选出差异表达基因.结果 与癌旁组织比较,原发病灶中存在着845个基因表达失调(9个上调,836个下调,P<0.01);癌症脑转移病灶中存在着813个基因表达失调(229个上调,584个下调,P<0.01);与肺腺癌原发病灶比较,癌症脑转移病灶中存在着218个基因表达失调(130个上调,88个下调,P<0.01).结论 转录组测序可以同步、全面的获取组织样本中的基因表达信息;随着肺腺癌病灶发生脑部转移,差异基因呈表达上调的趋势.

Objective To analyze the differentially expressed genes (DEGs) among primary lung cancer,adjacent normal lung tissues and the metastatic brain tissues.Methods The primary lung cancer tissues,adjacent normal lung tissues and metastatic brain tissues were collected from one patient for RNA-Seq analysis.DEGseq was utilized to identify the DEGs.Results Compared with the normal lung tissues,there were 845 DEGs in primary lung cancer tissues (9 up-regulated and 836 down-regulated genes,P < 0.01);there were 813 DEGs in metastatic brain tissues (229 up-regulated and 584 down-regulated genes,P < 0.01);compared with primary lung cancer tissues,there were 218 DEGs in metastatic brain tissues (130 up-regulated and 88 down-regulated genes,P < 0.01).Conclusion RNA-Seq analysis is an effective approach for obtaining the gene expression profile of tissue samples;the DEGs showed an up-regulation trend along with the occurrence of primary lung cancer metastasis to the brain.
