目的 观察CXC趋化因子受体5(CXCR5)在结直肠癌组织的表达,及其对结肠癌细胞株LoVo的侵袭转移的影响.方法 采用免疫组织化学链霉菌抗生物素蛋白-过氧化物酶(SP)法检测167例相对应的结直肠癌、癌旁肠黏膜组织,87例结直肠腺瘤标本中CXCR5的表达.通过RNA干扰(RNAi)技术构建4条CXCR5短发卡RNA(shRNA)表达载体,慢病毒法感染至高表达CXCR5的kVo细胞,用Western blot法检测shRNA对CXCR5的干扰效率.应用Transwell侵袭小室方法观察干扰CXCR5基因后,LoVo细胞迁移及侵袭能力的变化.结果 结直肠癌组织中CXCR5的阳性表达率为46.7%,明显高于癌旁肠黏膜组织(0.6%)及结直肠腺瘤组织(22.5%),差异有统计学意义(P<0.05).在有淋巴结转移及远处转移的患者中阳性率(55.4%与61.4%)明显较高(P<0.05).LoVo细胞转染CXCR5 shRNA,应用Western blot检测其干扰效率,发现sh1、sh2和sh3能够降低CXCR5表达,其中sh1和sh3效率较高.CXCR5 shRNA敲低后LoVo细胞侵袭能力下降(t=15.824,P<0.01).Transwell侵袭小室实验证实CXCR5 shRNA干扰后LoVo细胞侵袭能力明显下降.结论 CXCR5在结直肠癌组织中呈高表达,CXCR5在结直肠癌的侵袭和转移过程中发挥重要作用.

Objective To investigate the clinical significance of CXC chemokine receptor 5 (CXCR5) in the pathogenesis and development of colorectal cancer,and its effect on invasion and metastasis of colon cancer cell line Lovo.Methods Immunohistochemical streptavid-in-peroxidase (SP) method was used to detect the expression of CXCR5 in 167 cases of colon cancer,corresponding paracancerous normal intestinal mucosa,and 87 cases of colon adenoma specimens.Four CXCR5 short hairpin RNA (shRNA) expression vectors were constructed by RNA interference (RNAi) technique,and transfected into LoVo cells with high expression of CXCR5 by the lentiviral method.The interference efficiency of shRNA on CXCR5 was detected by Western blotting.The Transwell invasion chamber method was used to observe the changes of migration and invasion ability of LoVo cells after interference of the CXCR5 gene.Results The protein levels of CXCR5 in colorectal cancer (46.7%) was up-regulated as compared with the corresponding paracancerous tissues (0.6%) and adenoma (22.5%,P <0.05).The positive rate of CXCR5 expression in patients with lymph node metastasis (55.4%) and distant metastasis (61.4%) was significantly higher (P < 0.05).LoVo cells were transfected with the shRNA and the interference efficiency was detected by Western blotting.Shl,sh2 and sh3 could knock down CXCR5,and the interference efficiency of sh1 and sh3 was higher.The invasion ability of LoVo cells was decreased after CXCR5 knockdown by shRNA (t =15.824,P < 0.01).The invasion ability of LoVo cells after shRNA interference of CXCR5 was significantly decreased.Conclusion CXCR5 is highly expressed in colorectal cancer tissues,and it may play an important role in colorectal cancer invasion and migration.CXCR5 may become a new target for treatment of colorectal cancer.
