目的 探讨缺氧诱导因子-1α(HIF-1 α)和血管内皮生长因子(VEGF)在胃癌组织中的表达及其与肿瘤细胞增殖的关系.方法 采用免疫组织化学链霉菌抗生物素蛋白-过氧化物酶(SP)法检测58例胃癌组织和癌旁组织中HIF-1α和VEGF的表达,并探讨它们与胃癌临床病理特征的关系,同时用流式细胞仪检测S期细胞比例(SPF).结果 HIF-1α在胃癌组织中阳性表达率为62.07% (36/58),在癌旁组织中阳性表达率为18.97%(11/58),两者差异有统计学意义(x2=6.49,P<0.05);淋巴结转移阳性组的阳性表达率为71.88% (23/32),淋巴结转移阴性组的阳性表达率为26.92(7/26,x2 =6.21,P<0.05).VEGF在胃癌组织中阳性表达率为65.52%(38/58),在癌旁组织中阳性表达率为22.41% (13/58),两者差异有统计学意义(x2=5.32,P<0.05);淋巴结转移阳性组的阳性表达率为68.75% (22/32),淋巴结转移阴性组的阳性表达率为30.77(8/26,x2=5.43,P<0.05).胃癌组织中HIF-1α和VEGF的表达与SPF值明显相关;胃癌组织中HIF-1 α和VEGF的表达呈正相关.结论 胃癌组织中HIF-1α和VEGF的表达升高,且两者的表达均与淋巴结转移相关;HIF-1α和VEGF可以促进胃癌肿瘤细胞的增殖.

Objective To explore the expression of hypoxia inducible factor-1 α (HIF-1α) and vascular endothelial growth factor (VEGF) in gastric carcinoma and its relationship with tumor cell proliferation.Methods The expression of HIF-1α,and VEGF in 58 cases of gastric carcinoma tissues and 58 cases of paracancerous tissues was detected by streptavidin-peroxidase immunohistochemical staining,and S phase fraction (SPF) was assessed by flow cytometry.Results In 58 cases of gastric carcinoma tissues,the positive rate of HIF-1α was 62.07% (36/58) and that of VEGF was 65.52% (38/58) respectively,significantly higher than that in paracancerous tissues.The positive rate of HIF-1α and VEGF in gastric carcinoma tissues with lymph node metastasis was higher than that without lymph node metastasis [71.88% (23/32) vs.26.92% (7/26);x2 =6.21,P <0.05) and [68.75% (22/32) vs.30.77% (8/26);x2 =5.43,P < 0.05].There was a strong association between the expression of HIF-1 α and VEGF with SPF (t =6.826;t =5.348,P < 0.01).The positive expression of HIF-1 α showed a positive correlation with the positive expression of VEGF.Conclusion The raised expression of HIF-1 α and VEGF was detectable in gastric carcinoma tissues.The expression of HIF-1α and VEGF was correlated with lymph node metastasis.HIF-1 α and VEGF could make the development by promoting the proliferation of gastric carcinoma cells.
