目的 通过对趋化因子受体7(CXCR7)的基因干扰,观察其对人结肠癌细胞株LoVo的侵袭、转移能力的影响.方法 将靶向CXCR7基因的携带短发夹RNA(shRNA)表达的质粒转入LoVo细胞株,以体外侵袭实验观察LoVo细胞侵袭能力的改变;将36只裸鼠分成3组,行种植成瘤肝转移实验观察LoVo细胞转移能力的改变.结果 体外实验结果显示CXCR7-shRNA干扰组LoVo细胞穿膜数[(51.70 ±6.89)个]较空白对照组[(94.30±7.40)个]减少(P<0.05);动物实验结果显示3组裸鼠的中位生存时间分别为:CXCR7-shRNA干扰组52 d、空白对照组30 d、阴性对照组32 d.CXCR7-shRNA干扰组较空白对照组生存率明显提高(P<0.05),肝脏表面的转移瘤结节数较空白对照组明显减少(P>0.05).空白对照组与阴性对照组比较差异无统计学意义(P>0.05).结论 shRNA干扰技术沉默CXCR7的表达能抑制人结肠癌细胞LoVo的侵袭和转移能力.

Objective To investigate the effects of chemokine receptor 7 (CXCR7) gene on the proliferation and migration of LoVo cells of human colon cancer cell line.Methods The expression vector with short hairpin RNA (shRNA) targeting CXCR7 was transfected into LoVo cells.The expression of knockdown CXCR7 gene was confirmed by Western blotting.To investigate the effects of the expression of CXCR7 on proliferation and migration of LoVo cells,Transwell migration assay was used in vitro,and tumor xenografts with liver metastasis model were performed in vivo,respectively.Results The number of LoVo cells migrating across the polycarbonate membrane in the CXCR7-shRNA transfection group (51.70 ±6.89) was significantly less than that in the control groups (94.30 ±7.40,P <0.05).The median survival time in CXCR7-shRNA transfection group,blank contorl group and negative control group was respectively 52,30,and 32 days.The survival rate in CXCR7-shRNA transfection group was significantly higher than in blank contorl group (P < 0.05).The amount of the tumor nodules on the liver surface in nude mice of CXCR7-shRNA transfection group was dramatically reduced (P > 0.05).No significant difference was found between blank contorl group and negative control group (P > 0.05).Conclusion RNA interference targeting CXCR7 gene can inhibit the proliferation and migration of LoVo cells.
