目的 观察甲状腺乳头状癌(PTC)及癌旁正常组织中卷曲螺旋结构域67(CCDC67)基因转录水平及其启动子甲基化状态.方法 采用反转录-聚合酶链反应(RT-PCR)、焦磷酸测序的方法检测60例PTC及其相对应的癌旁组织中CCDC67的转录以及基因启动子甲基化.结果 癌旁组织中均出现CCDC67 mRNA的表达(100.0%,60/60),而在PTC中CCDC67基因mRNA表达明显下降,为40.0%(24/60)(P<0.05).在PTC组织Ⅰ~Ⅱ期与Ⅲ~ Ⅳ期中,CCDC67 mRNA表达率分别为68.0%(17/25)、20%(7/35),差异有统计学意义(P<0.01).在病理分级Ⅰ级和Ⅱ级中,CCDC67 mRNA的表达率分别为60.8%(14/23)和27.0% (10/37),差异有统计学意义(P<0.01).在有淋巴结转移的PTC中CCDC67 mRNA的表达率为25.0% (9/36),在无淋巴结转移的PTC中CCDC67 mRNA的表达率为62.5% (15/24),差异有统计学意义(P<0.01).焦磷酸测序结果显示60例PTC组织中只有2例出现CCDC67基因的甲基化,且与CCDC67基因mRNA的表达无明显相关.结论 CCDC67在PTC的发生发展过程中可能扮演着重要角色.CCDC67基因的甲基化与mRNA表达的异常无明显相关.

Objective To observe the mRNA expression and promotor methylation of coiled coil domain 67 (CCDC67) gene in papillary thyroid carcinoma (PTC).Methods Reverse transcription-polymerase reaction (RT-PCR) and pyrosequencing were used to detect the mRNA and promotor methylation of CCDC67 gene in 60 human PTC specimens and their matched adjacent normal tissues.Results All cases had expression of CCDC67 mRNA in adjacent normal tissues,but the expression of CCDC67 mRNA was down-regulated significantly,only 40.0% (24/60) in 60 cases of thyroid carcinoma was positive for the CCDC67 mRNA expression.The CCDC67 mRNA expression was correlated with the tumor TNM stage,pathological grade and lymph node metastasis (P < 0.05).Pyrosequencing results showed that there were only 2 methylated cases in 60 cases of PTC.Furthermore there was no distinct correlation between the mRNA and promotor methylation of CCDC67 gene (P < 0.01).Conclusion CCDC67 may play an important role in the occurrence and development of PTC.There was no distinct correlation between the mRNA and promotor methylation of CCDC67 gene.
