目的 探讨血卟啉单甲醚光动力治疗(HMME-PDT)处理后人胶质瘤U251细胞长链非编码RNA(lncRNA)表达谱的变化.方法 使用HMME-PDT处理人胶质瘤U251细胞,提取RNA,体外反转录制备并标记为双链cDNA,在标准条件下与含有33 045条lncRNA及30215条mRNA的芯片杂交,计算机扫描荧光信号图像,分析lncRNA和mRNA表达谱的变化.结果 在HMME-PDT处理的人U251细胞中,lncRNA表达上调l 121条,表达下调1 025条;mRNA表达上调1 111条,表达下调935条(变化>2倍且P< 0.05).结论 HMME-PDT处理后人胶质瘤细胞lncRNA/mRNA发生明显变化,lncRNA与光动力处理后抗原呈递元件(APM)表达变化密切相关.

Objective To study hematoporphyrin monomethyl ether photodynamic therapy (HMME-PDT) treatment posterity glioma U251 cell lines long chain noncoding RNA (lncRNA) expression changes,and the results are analyzed.Methods Using HMME-PDT treatment of glioma U251 cell lines,preparation and extraction of RNA reverse transcription in vitro markers for double chain cDNA,under the condition of the standard and contains 33 045 long non-coding RNAs (lncRNA) and 33 045 mRNA chip hybridization,computer scanning fluorescent signal image,analyzing the change of the lncRNA/mRNA expression.Results In HMME-before and after treatment were treated with PDT U251 cells,raised 1 121 lncRNA,express by 1 025;Raised 1 111 mRNA,cut in 935 (>2 times and P< 0.05).Conclusion HMME-PDT treatment posterity glioma cells IncRNA has obvious changes,antigen processing machinery (APM) lncRNA antigen-presenting after PDT treatment related protein expression changes have close relationship.
