目的:观察清毒汤对实验性重症肝损伤大鼠内毒素( endotoxin,ET)、肿瘤坏死因子α(tumor necrosis factor alpha,TNF-α)以及核转录因子κB(nuclear factor kappa B,NF-κB)表达的影响,探讨该方治疗慢性重型肝炎内毒素血症的作用机制。方法建立硫代乙酰胺( thioacetamide,TAA)致实验性大鼠重症肝损伤模型,给予清毒汤干预后,检测大鼠ET、TNF-α以及NF-κB表达的水平。结果与正常组比较,模型组大鼠血清ET、TNF-α以及NF-κB蛋白含量水平均显著升高(P<0.01);与模型组比较,清毒汤中、低剂量组和乳果糖组 ET、TNF-α以及 NF-κB 蛋白含量水平显著降低(P<0.01),清毒汤高剂量组除ET外各指标均明显改善(P<0.05);与乳果糖组比较,清毒汤中剂量组ET及TNF-α水平显著降低(P<0.01),低剂量组ET及TNF-α水平无明显差异(P>0.05),高剂量组ET及TNF-α水平明显高于乳果糖组(P<0.05)。结论清毒汤对实验性肝损伤大鼠模型具有降低ET、TNF-α以及NF-κB蛋白水平的作用,尤其中剂量组效果最为明显,推测其分子机制可能为通过抑制NF-κB因子的表达从而达到保肝目的。

목적:관찰청독탕대실험성중증간손상대서내독소( endotoxin,ET)、종류배사인자α(tumor necrosis factor alpha,TNF-α)이급핵전록인자κB(nuclear factor kappa B,NF-κB)표체적영향,탐토해방치료만성중형간염내독소혈증적작용궤제。방법건립류대을선알( thioacetamide,TAA)치실험성대서중증간손상모형,급여청독탕간예후,검측대서ET、TNF-α이급NF-κB표체적수평。결과여정상조비교,모형조대서혈청ET、TNF-α이급NF-κB단백함량수평균현저승고(P<0.01);여모형조비교,청독탕중、저제량조화유과당조 ET、TNF-α이급 NF-κB 단백함량수평현저강저(P<0.01),청독탕고제량조제ET외각지표균명현개선(P<0.05);여유과당조비교,청독탕중제량조ET급TNF-α수평현저강저(P<0.01),저제량조ET급TNF-α수평무명현차이(P>0.05),고제량조ET급TNF-α수평명현고우유과당조(P<0.05)。결론청독탕대실험성간손상대서모형구유강저ET、TNF-α이급NF-κB단백수평적작용,우기중제량조효과최위명현,추측기분자궤제가능위통과억제NF-κB인자적표체종이체도보간목적。
Objective To observe the effects of Qingdu decoction on the expressions of endotoxin ( ET ) , Tumor necrosis factor alpha ( TNF-α) and Nuclear factor kappa B ( NF-κB ) in rats with severe liver injury and explore the mechanism of Qingdu decoction on treating Chronic severe hepatitis complicated with endotoxemia. Methods The severe liver injury rat model was induced by Thioacetamide ( TAA ) , and treated with Qingdu decoction. And then the blood level of ET, TNF-α and NF-κB were measured. Results The expression levels of ET, TNF-αand NF-κB in model group were significantly increased when compared to that of in normal group (P<0. 01 ). Compared to the model group, the expression levels of ET, TNF-α and NF-κB were decreased significantly in the middle and low dose Qingdu decoction groups and Lactulose group (P<0. 01 ). All indexes except ET in the high dose Qingdu decoction group were decreased significantly (P<0. 05 ). Compared to lactulose group, the expressions of ET and TNF-αin the middle dose Qingdu decoction group were decreased significantly(P<0. 01 ), while the expressions of ET and TNF-α in the low dose group showed no significantly difference (P>0. 05 ). The expressions of ET and TNF-α in the high dose group were significantly higher than the Lactulose group ( P<0. 05 ) . Conclusions Qingdu decoction can improve liver injury and decrease ET, TNF-α and NF-κB expression levels in rats with hepatic injury complicated with endotoxemia, especially in the middle dose group. The mechanism could be related to the inhibition of NF-κB activity.
