目的:评价黄连和大黄联合头孢他啶体内外抗菌作用。方法制备病原菌悬液,采用棋盘滴定法测定大黄、黄连素联合头孢他啶测定最低抑菌浓度(MIC),并取昆明小鼠进行体内实验,制得金黄色葡萄球菌败血症模型,灌胃0.2 ml配比不同浓度大黄、黄连联合头孢他啶药液,计算存活率、有效剂量ED50及其95%可信区间。结果大黄、黄连联合头孢他啶,金黄色葡萄球菌、大肠埃希菌、铜绿假单胞菌MIC、FIC指数低于单独应用头孢他啶,作用效果为协同或相加;低剂量、总剂量、高剂量,头孢他啶联合大黄、黄连存活小鼠均高于单独应用头孢他啶,药物联合大黄、黄连ED50低于单独应用头孢他啶。结论黄连和大黄联合头孢他啶有助于增强体内外抗菌作用,具有协同共增强效用。

목적:평개황련화대황연합두포타정체내외항균작용。방법제비병원균현액,채용기반적정법측정대황、황련소연합두포타정측정최저억균농도(MIC),병취곤명소서진행체내실험,제득금황색포도구균패혈증모형,관위0.2 ml배비불동농도대황、황련연합두포타정약액,계산존활솔、유효제량ED50급기95%가신구간。결과대황、황련연합두포타정,금황색포도구균、대장애희균、동록가단포균MIC、FIC지수저우단독응용두포타정,작용효과위협동혹상가;저제량、총제량、고제량,두포타정연합대황、황련존활소서균고우단독응용두포타정,약물연합대황、황련ED50저우단독응용두포타정。결론황련화대황연합두포타정유조우증강체내외항균작용,구유협동공증강효용。
Objective To evaluate the antibacterial effect in vitro and in vivo of coptis chinensis and rhubarb combined with ceftazidime.Methods Pathogenic bacteria suspension was prepared by checkerboard titration method for the determination of rhubarb,berberine combined with ceftazidime checked minimal inhibitory concentration (MIC). Kunming mice were taken in vivo experiment,and making staphylococcus aureus sepsis model,intragastric administration of 0.2 ml ratio of different concentrations of rhubarb,coptis chinensis combined with ceftazidime liquid and calculated the survival rate, effective dose(ED50)and 95% confidence interval.Results Rhubarb,coptis chinensis combined with ceftazidime caused Staphylococcus aureus, Escherichia coli,Pseudomonas aeruginosa of MIC, FIC index less than ceftazidime alone,the effect was synergistic or additive. Low dose,the total dose,high dose of Rhubarb, coptis chinensis combined with ceftazidime,surviving mice were higher than ceftazidime alone. ED50 of drug combination rhubarb,coptis chinensis was less than ceftazidime alone.ConclusionThe ceftazidime combined with coptis chinensis and rhubarb was helpful to enhance the antibacterial effect in vitro and in vivo,and it had synergistic effect.
