目的:检测B7家族B7-H5以及CD28H在直肠癌组织中的表达情况。方法收集来我院治疗的78例结直肠癌患者的组织样本,通过免疫组化方法,以及实时定量PCR方法检测其B7-H5/CD28H表达情况。结果直肠癌组织部分B7-H5以及CD28H mRNA表达水平均高于癌旁组织(P<0.05)与正常组织(P<0.05),癌组织分组比较无显著差异(P>0.05),癌旁组织与正常组织无显著差异(P>0.05)。免疫组化方法检测B7-H5蛋白表达水平显示,癌旁组织,正常组织与癌组织阳性率分别为83.3%,74.3%和23.1%,与癌组织比较均具有差异(P<0.05),癌组织分组间无显著差异(P>0.05),癌旁组织与正常组织无显著差异(P>0.05)。结论不同于B7家族其他成员,结直肠癌组织中B7-H5/CD28H的表达量下降,可能与免疫逃逸有关,因此可以成为一个新的检测结直肠癌的标志物。

목적:검측B7가족B7-H5이급CD28H재직장암조직중적표체정황。방법수집래아원치료적78례결직장암환자적조직양본,통과면역조화방법,이급실시정량PCR방법검측기B7-H5/CD28H표체정황。결과직장암조직부분B7-H5이급CD28H mRNA표체수평균고우암방조직(P<0.05)여정상조직(P<0.05),암조직분조비교무현저차이(P>0.05),암방조직여정상조직무현저차이(P>0.05)。면역조화방법검측B7-H5단백표체수평현시,암방조직,정상조직여암조직양성솔분별위83.3%,74.3%화23.1%,여암조직비교균구유차이(P<0.05),암조직분조간무현저차이(P>0.05),암방조직여정상조직무현저차이(P>0.05)。결론불동우B7가족기타성원,결직장암조직중B7-H5/CD28H적표체량하강,가능여면역도일유관,인차가이성위일개신적검측결직장암적표지물。
Objective To detect the B7 family B7-H5 and CD28H expression in colorectal cancer tissue.MethodsColected 78 cases of patients with colorectal cancer tissue samples from our hospital,used immunohistochemical method,and real-time quantitative polymerase chain reaction (PCR) to detect the B7-H5/CD28H expression.Results Rectal cancer tissue section B7-H5 and CD28H mRNA expression levels were higher than tissue adjacent to carcinoma(P< 0.05)and normal group(P< 0.05),the cancer group was no significant difference(P> 0.05).Tissue adjacent to carcinoma and normal group there was no significant difference(P> 0.05). Immunohistochemical method detected B7-H5 protein expression levels,according to the tissue adjacent to carcinoma and normal tissues and the cancerous tissue positive rate were 83.3%,74.3% and 23.1%,compared with the cancer had significant difference(P< 0.05),carcinoma group(P> 0.05), no significant differences between tissue adjacent to carcinoma and normal group there was no significant difference(P> 0.05).Conclusion Different from other members of the family of B7,colorectal cancer tissues decreased the expression of B7-H5/CD28H,may be associated with immune escape,so it can become a new detect colorectal cancer markers.
