利用大肠杆菌的原核表达体系高效表达SEB基因,获得了SEB重组蛋白(rSEB);通过优化SEB较好发挥免疫效果时的免疫剂量,选取5μg/只SEB分别协同AIV-H9/NDV二联灭活苗和AIV-H5灭活疫苗对2周龄的艾维因肉鸡进行免疫,定期测定HA抗体滴度及淋巴细胞活性以评价SEB的免疫增强效果。结果表明,SEB免疫组的NDV和AIV的HA抗体效价显著高于疫苗对照组(P﹤0.05),rSEB中剂量组(5μg/只)佐剂效果明显(P﹤0.05);SEB协同免疫组的淋巴细胞活性在免疫后第15天达到峰值,随后逐渐下降,至第25天与疫苗对照组相比,差异仍然显著(P﹤0.05)。

Staphylococcus aureus enterotoxin B (SEB) is a bacterial superantigen (SAg) with a strong immune activation capa-bilities and leading to lots of cytokines release. SEB has the potential to become an immune adjuvant. To explore its use in veterinary area, recombinant (rSEB) or commercial SEB (sSEB) was injected with commercial AIV-H9/NDV or AIV-H5 inactive vaccines in 2-week-old avian broilers respectively. The results showed rSEB orsSEB could enhance the immune re-sponse by testing the antibody titers and lymphocyte activity. Statistical analysis showed that anti-HA titers of SEB+vaccine group were significantly higher than the control group (P﹤0.05), and that of 5μg-rSEB+vaccine group were significantly dif-ferent with those of other concertration rSEB groups (P﹤0.05). Furthermore, chicken in 5ng-rSEB+vaccine group had better immune response. The lymphocyte activity peaked 15 d after immunization, and the anti-HA antibody titer were about 2.5 times than only vaccine group (P﹤0.05). These significant differences maintained up to more than 25 days after vaccine. In conclusion, our results indicated that the appropriate dose of SEB collaborated with vaccine could enhance the immune re-sponse to the vaccine in both humoral and cellular immunity. Moreover, SEB was predicted as a new and effective vaccine adjuvant with a good prospect in the future.
