目的:建立测定大鼠血浆中京尼平苷酸的 UHPLC?MS/MS 方法,研究盐炙对杜仲中京尼平苷酸药代动力学的影响。方法色谱柱:BEH?C18(100 mm×2.1 mm,1.7μm);流动相:0.1%甲酸水?乙腈梯度洗脱;流速:0.3 mL/min。结果京尼平苷酸在生品和盐炙品组大鼠体内的药动学参数分别为:AUC(0?t)(1547.18±272.28)、(2120.694±664.532)ng· h/mL;AUC(0?∞)(1564.42±273.97)、(2145.61±659.983)ng·h/mL;Cmax (517.59±51.24)、(733.292±261.34)ng/mL;MRT(2.68±0.11)、(2.551±0.08)h;T1/2(1.37±0.08)、(1.43±0.17)h;Tmax (1.52±0.1)、(1.51±0.1)h。结论杜仲盐炙后有助于促进京尼平苷酸的吸收,一定程度上增加了其生物利用度。

ABSTRACTOBJECTIVE To establish UHPLC?MS/MS method to determine geniposidic acid in rat plasma and to investigate the effect of salt processing on pharmacokinetics of geniposidic acid in Eucommia ulmoides Oliver.METHODS Chromato-graphic columnBEH?C18100 mm×2.1 mm1.7 μmthe mobile phaseacetonitrile?0.1% formic acid aqueous solution.
