目的:观察蒙药额尔敦·乌日勒在大鼠脊髓不完全损伤后的神经修复作用,探讨其对Nogo -A表达的调节作用。方法把成年Wistar大鼠48只随机分为损伤组(A组)和治疗组(B组),每组24只。利用改良Allen’s方法建立脊髓不完全损伤模型。损伤后用额尔敦·乌日勒对B组进行灌胃治疗。将A、B组分为3、7、14 d 3个时间点,每个时间点8只。分别在3个时间点对A、B组大鼠的运动功能进行BBB评分,评分完毕处死动物并取受损节段脊髓行免疫组化染色观察Nogo-A在脊髓组织中的分布情况,同时进行阳性细胞计数,并作比较。结果 A、B组大鼠的运动功能在损伤后都有不同程度的恢复,B组7 d和14 d的BBB评分明显高于A组对应时间点,差异有统计学意义( P<0.05)。Nogo -A蛋白在B组的表达量显著低于A组,差异有显著性( P<0.05)。结论大鼠脊髓损伤后Nogo-A表达明显升高,额尔敦·乌日勒具有抑制Nogo-A表达的作用。

목적:관찰몽약액이돈·오일륵재대서척수불완전손상후적신경수복작용,탐토기대Nogo -A표체적조절작용。방법파성년Wistar대서48지수궤분위손상조(A조)화치료조(B조),매조24지。이용개량Allen’s방법건립척수불완전손상모형。손상후용액이돈·오일륵대B조진행관위치료。장A、B조분위3、7、14 d 3개시간점,매개시간점8지。분별재3개시간점대A、B조대서적운동공능진행BBB평분,평분완필처사동물병취수손절단척수행면역조화염색관찰Nogo-A재척수조직중적분포정황,동시진행양성세포계수,병작비교。결과 A、B조대서적운동공능재손상후도유불동정도적회복,B조7 d화14 d적BBB평분명현고우A조대응시간점,차이유통계학의의( P<0.05)。Nogo -A단백재B조적표체량현저저우A조,차이유현저성( P<0.05)。결론대서척수손상후Nogo-A표체명현승고,액이돈·오일륵구유억제Nogo-A표체적작용。
Objective To investigate the effects of mongolian medicine Eerdun Wurile on the nerve regen-eration in rats with acute incomplete spinal cord injury and it ’s regulating effect on expression of Nogo -A . Method Rat model of spinal cord injury was established by modified Allen ’s method .Forty -eight adult Wis-tar rats were divided into the spinal cord injury group (group A ,n=2 4 ) and group B ( n=2 4 ) treated with Eerdun Wurile at random after spinal cord injury .The rats in group B was treated with Eerdun Wurile (1 g/kg) once a day by gavage .At 3 d ,7 d and 1 4 d after operation ,motor functions of the rat hind limbs were determined by the BBB (Basso Beattie and Bresnahan ) ,and the rats in each group were killed at 3 d ,7 d and 1 4 d post opera-tion for historygcial examination .Morphologic changes of spinal neurons were evaluated by HE staining and im-munohistochemistry ,separately .Results It was observed that at 3 d ,7 d and 1 4 d after operation ,the motor functions of the rat hind limbs about the acute spinal cord injury in each group were recovered .The expression levels of Nogo -A were significantly decreased in group B(P<0 .05 ) .Conclusion The expression of Nogo -A was markedly increased in rats with acute spinal cord injury ,and early use of Eerdun Wurile may inhibit the ex-pression of Nogo -A .
