目的:应用蛋白质组学双向电泳和质谱技术,开展强直性脊柱炎湿热痹阻证证候学研究。方法对强直性脊柱炎湿热痹阻证、寒湿痹阻证、正常对照共3组各30例血清,分别进行双向凝胶电泳,软件分析获取湿热痹阻证组和其他2组的差异蛋白质点,二级质谱鉴定获得差异蛋白。应用 Western blot 验证差异蛋白。结果获得7个湿热痹阻证的共同差异蛋白点,经质谱鉴定和 SwissProt 数据库检索,获得 Fibulin?1、二肽基肽酶1、激肽原1、白细胞介素17F 突变型、甘露糖结合蛋白 C 等5个强直性脊柱炎湿热痹阻证差异蛋白,涉及胞外基质蛋白、蛋白酶类、凝血因子、细胞因子、转运蛋白等。Western blot 验证甘露糖结合蛋白 C 的表达结果与蛋白组学检测结果一致。结论这些蛋白可能涉及强直性脊柱炎湿热痹阻证的发病和病理过程,为进一步研究湿热痹阻证实质提供了依据。

목적:응용단백질조학쌍향전영화질보기술,개전강직성척주염습열비조증증후학연구。방법대강직성척주염습열비조증、한습비조증、정상대조공3조각30례혈청,분별진행쌍향응효전영,연건분석획취습열비조증조화기타2조적차이단백질점,이급질보감정획득차이단백。응용 Western blot 험증차이단백。결과획득7개습열비조증적공동차이단백점,경질보감정화 SwissProt 수거고검색,획득 Fibulin?1、이태기태매1、격태원1、백세포개소17F 돌변형、감로당결합단백 C 등5개강직성척주염습열비조증차이단백,섭급포외기질단백、단백매류、응혈인자、세포인자、전운단백등。Western blot 험증감로당결합단백 C 적표체결과여단백조학검측결과일치。결론저사단백가능섭급강직성척주염습열비조증적발병화병리과정,위진일보연구습열비조증실질제공료의거。
ABSTRACTOBJECTIVE Proteomic technique was applied to analyze ankylosing spondylitis AS patients with damp?heat arthralgia spasm syndrome.METHODS Serum proteins of AS patients with damp?heat arthralgia spasm syndrome groupn=30 was compared with that of normal control group n = 30 and AS patients with cold?dampness arthralgia spasm syn-drome groupn =30 respectively by two?dimensional gel electrophoresisthe common differential protein spots of damp?heat arthralgia spasm syndrome were analyzed by the second order MS.The validation of the differential protein was performed u-sing Western blot.RESULTS Seven kinds of common differential protein spots were selected and five kinds of proteins were i-dentified by mass spectrometry.They were fibulin 1dipeptidy lpeptidase 1kininogen 1mutant IL 17Fmannose?binding protein C and related to extracellular matrix proteinproteasecoagulation factorcytokinetransport proteinand so on.Re-sults of Western blot were consistent with the proteomic results.CONCLUSION These proteins are implicated in the patho-genesis and pathological processes of AS with damp?heat arthralgia spasm syndromeand provide a basis for further studies of the essence of the syndrome in traditional Chinese medicine.
