泛素蛋白酶体系统是广受关注的抗肿瘤药物靶标,细胞内蛋白酶体活性变化的实时检测对抗肿瘤药物筛选及作用机理研究具有重要意义。本文构建了稳定表达GFPu蛋白的人前列腺癌PC-3细胞系,并建立了对GFPu蛋白荧光图像的定量分析方法。该方法可以在不影响细胞正常生命过程,不加入荧光底物的条件下,实现对细胞内蛋白酶体活性变化的实时、动态、定量监控。

The ubiquitin-proteasome system is a widely concerned target of anticancer drugs. Real-time detection of intracellular proteasome activity is of great significance for the screening and mechanism study of anticancer drugs. In this paper,Human prostate cancer PC-3 cell lines which stably expressing GFPu was constructed. Furthermore,a quantitative analysis method for the GFPu fluorescence images had been established, which can realize real-time, dynamic, quantitative monitoring of proteasome activity change without affecting the normal cell life process or adding of fluorescent substrate.
