目的 研究北五味子木脂素( Schisandra Chinensis Baill Lignans,SCL)抗焦虑的作用及机制.方法 采用明暗箱试验和高架十字迷宫试验,观察北五味子木脂素低、中、高剂量(35,70,140 mg/kg)灌胃给药7 d后对小鼠行为的影响;另设空白对照组(给予生理盐水)和地西泮(2 mg/kg)阳性对照组.用酶联免疫吸附法( ELISA)测定小鼠血清γ-氨基丁酸( GABA)的含量.结果 明暗箱试验和高架十字迷宫试验结果显示:北五味子木脂素中、高剂量组具有显著的抗焦虑作用(P<0. 05),并呈剂量依赖关系,这种抗焦虑作用可被氟马西尼所拮抗.酶联免疫吸附试验显示:中、高剂量组小鼠血清中γ-氨基丁酸( GABA)的含量显著增高.结论 北五味子木脂素有显著的抗焦虑作用,其机制可能是通过γ-氨基丁酸受体(γ-aminobutyric acid,GABA-R)而发挥作用.

Objective To study the anti-anxiety function and mechanism of Schisandra chinensis Baill Lignans ( SCL) . Method Light-dark box test and elevated plus maze test were used to observe the influence of Schisandra chinensis Baill Lignans on mice behaviors administrated intragastrically at low,medium and high dose (35,70 and 140 mg/kg) after 7 days. The blank control group and the positive control group were given normal saline and diazepam(2 mg/kg),respectively. Enzyme-linked immunoabsorbent assay(ELISA) was used for the detection of gamma aminobutyric acid(GABA) in mice serum. Results Light-dark box test and elevated plus maze test results showed that SCL had significant anti-anxiety function at medium and high dose(P<0. 05) with a dose-response relationship. And this kind of anti-anxiety effect could be antagonized by flumazenil. ELISA experiments showed that serum gamma aminobutyric acid ( GABA ) levels of the mice in the medium and high dose group increased significantly. Conclusion Schisandra chinensis Baill Lignans showed significantly anti-anxiety effect,its mechanism may be related with gamma-aminobutyric acid receptor( gamma aminobutyric acid, GABA-R) .
