目的 探讨应用D-半乳糖( D-gal)建立小鼠肝脏损伤模型的方法.方法 将20只小鼠随机分为两组,正常对照组应用生理盐水20 mL/( kg·d-1 )皮下注射,模型组应用D-半乳糖按500 mg/( kg·d-1 ) 皮下注射.造模完成后采用比色法测定血清AST,ALT及GST;放免法测定血浆PGE2,TNF及IL-6;Western blot方法检测肝组织COX-2表达.通过光镜观察肝组织病理变化.结果 血浆ALT,AST及GST模型组与正常对照组比较有明显升高(P<0. 01);模型组与正常对照组比较血浆PGE2,TNF及IL-6升高明显,差异具有统计学意义(P<0. 01);病理切片显示肝脏结构呈现病理改变.结论 应用D-半乳糖可以成功制作小鼠肝脏损伤模型.

Objective To explore the application of D-galactose in producing liver injury model in mice. Method 20 mouse were randomly divided into 2 groups. The normal control group was injected saline 20 mL/( kg · d-1 ) subcutaneously, and the model group was administrated D-galactose 500 mg/( kg · d-1 ) subcutaneously. After the model was established,colorimetric method was used to determine serum AST,ALT and GST,plasma concentrations of PGE2 TNF and IL-6 were measured by radio immunity, and the expression of COX-2 was detected by Western blot. Meanwhile, pathological change of the liver tissue was observed through light microscope. Results Compared with the normal control group, serum ALT, AST and GST of the model group increased significantly(P<0. 01),plasma PGE2,TNF and IL-6 also increased obviously with statistical significance(P<0. 01). Pathological section indicated clear changes in liver tissue of the mice in model group. Conclusion D-galactose can successfully establish liver injury model in mice.
