目的:比较临床样本中肿瘤周围与肿瘤远端的脂肪组织来源的间充质干细胞生物学特征差异。方法流式细胞术鉴定两种来源脂肪间充质干细胞表面标志;Realtime-PCR检测两种来源脂肪间充质干细胞相关基因的表达变化。结果癌周脂肪组织中AMSCs表达更高的干细胞相关基因Oct4,Nanog,Rex1;生长因子IGF,HGF;趋化因子SDF-1。结论肿瘤微环境影响下的脂肪间充质干细胞可能增强结肠肿瘤细胞的侵袭能力。
目的:比較臨床樣本中腫瘤週圍與腫瘤遠耑的脂肪組織來源的間充質榦細胞生物學特徵差異。方法流式細胞術鑒定兩種來源脂肪間充質榦細胞錶麵標誌;Realtime-PCR檢測兩種來源脂肪間充質榦細胞相關基因的錶達變化。結果癌週脂肪組織中AMSCs錶達更高的榦細胞相關基因Oct4,Nanog,Rex1;生長因子IGF,HGF;趨化因子SDF-1。結論腫瘤微環境影響下的脂肪間充質榦細胞可能增彊結腸腫瘤細胞的侵襲能力。
목적:비교림상양본중종류주위여종류원단적지방조직래원적간충질간세포생물학특정차이。방법류식세포술감정량충래원지방간충질간세포표면표지;Realtime-PCR검측량충래원지방간충질간세포상관기인적표체변화。결과암주지방조직중AMSCs표체경고적간세포상관기인Oct4,Nanog,Rex1;생장인자IGF,HGF;추화인자SDF-1。결론종류미배경영향하적지방간충질간세포가능증강결장종류세포적침습능력。
Objective To investigate the characteristic by which human adipose-derived mesenchymal stem cel s(hAMSCs)derived from fat tissue adjacent to carcinoma. Methods Flow cytometry analysis for MSC-specific phenotype of AMSCs in the two sources of tissue. The transcripts of stemness-associated genes in hAMSCs were determined by a qRT-PCR assay. Results AMSCs were able to induce AMSCs to produce malignance-related genes,such as Oct4,Nanog,Rex1,IGF,HGF and SDF-1. Conclusion These findings thus suggest that the crosstalk between the microenvironment of cancer cel s and AMSCs has an implication in colon cancer malignancy.
