目的:探讨重楼皂苷Ⅶ联合顺铂抑制荷瘤小鼠移植瘤生长的分子机制。方法 SKOV3细胞皮下接种NOD/SCID 小鼠,待移植瘤形成后,荷瘤小鼠随机分为4组(n=10):对照组、低剂量重楼皂苷Ⅶ组(LD 组)、中剂量重楼皂苷Ⅶ组(MD 组)、高剂量重楼皂苷Ⅶ组(HD 组)。15天后,分离肿瘤组织,测定肿瘤体积和重量及抑瘤率;TUNEL 染色和 Western blot 检测肿瘤细胞凋亡及其机制。结果在 HD 组中,肿瘤重量抑制率达57.98%,体积抑制率达55.11%,细胞凋亡率达42.39%,与对照组相比有统计学差异。经重楼皂苷作用后,凋亡相关基因 Bcl-2表达减少,Bax 蛋白表达增加;内质网相关基因 GRP78和 CHOP 表达均增加。结论重楼皂苷能有效促进肿瘤细胞凋亡,并且这种凋亡是由内质网应激介导的。

목적:탐토중루조감Ⅶ연합순박억제하류소서이식류생장적분자궤제。방법 SKOV3세포피하접충NOD/SCID 소서,대이식류형성후,하류소서수궤분위4조(n=10):대조조、저제량중루조감Ⅶ조(LD 조)、중제량중루조감Ⅶ조(MD 조)、고제량중루조감Ⅶ조(HD 조)。15천후,분리종류조직,측정종류체적화중량급억류솔;TUNEL 염색화 Western blot 검측종류세포조망급기궤제。결과재 HD 조중,종류중량억제솔체57.98%,체적억제솔체55.11%,세포조망솔체42.39%,여대조조상비유통계학차이。경중루조감작용후,조망상관기인 Bcl-2표체감소,Bax 단백표체증가;내질망상관기인 GRP78화 CHOP 표체균증가。결론중루조감능유효촉진종류세포조망,병차저충조망시유내질망응격개도적。
Objective To investigate the molecular mechanism of inhibiting of tumor growth in tumor-bearing mice by polyphyllin Ⅶ and cisplatin.Methods SKOV3 cells were inoculated subcutaneously into NOD/SCID mice,and the tumor-bearing mice were randomly divided into four groups (n = 10):the control group,low-dose of polyphyllin Ⅶgroup (group LD),medium-dose of polyphyllin Ⅶ group (group MD),high-dose of polyphyllin Ⅶ group (group HD). 15 days later,the tumor tissues were isolated and measured volume and weight to calculate the tumor inhibition rate;the tumor cell apoptosis and its mechanism were determined by TUNEL staining and Western blot.Results In group HD, the weight inhibition rate was 57.98%,volume inhibition rate was 55.11%,and the apoptotic rate was 42.39%,there was a significant difference compared with the control group.After treatment of polyphyllin Ⅶ,the expression of Bcl-2 was decreased and the expression of Bax was increased;the expression of GRP78 and CHOP,the endoplasmic reticu-lum-related gene,was increased too.Conclusion The polyphyllins could effectively promote the apoptosis of tumor cells via endoplasmic reticulum stress.
