目的 建立HPLC法测定山茱萸果核中5种有机酸(草酸、酒石酸、DL-苹果酸、柠檬酸和没食子酸)含量的方法. 方法 采用Kromasil C18 (250 mm × 4. 6 mm i. d. , 5 μmm)色谱柱,3% CH3 OH 0. 01 mol·L-1 KH2 PO4 (pH 3. 0)溶液做流动相,流速0. 5 mL·min-1,柱温:25 ℃,检测波长210 nm. 结果 在32 min内实现了5种有机酸的基线分离,各有机酸测定的精密度为0. 29% ~1. 41%(n=6),回收率为89. 6% ~102. 0%. 结论 所建立的方法简便、准确、重现性好,可用于山茱萸果核中5种有机酸含量的测定.

Objective To establish a quantitative HPLC method for determining five kinds of organic acids(oxalic acid, tartaric acid, DL-malic acid, citric acid and gallic acid) in cornus officinalis kernels. Methods A Kromasil C18 column(250 mm × 4. 6 mm i. d. , 5 μmm) was used with 3% CH3OH-0. 01 mol·L-1 KH2PO4(pH 3. 0)as the mobile phase at a flow rate of 0. 5 mL·min-1. The column temperature was 25℃ and the UV detection wavelength was 210 nm. Results The five organic acids were separated in 32 min. The average recoveries of the method were 89. 6% ~102. 0% with the relative standard deviation of 0. 29% ~1. 41%(n=6). Conclusion The method is simple, accurate, reproducible and practical for the detection of organic acids in cornus officinalis kernels.
