目的:探讨同源盒基因转录反义RNA( HOTAIR)调控乳腺癌细胞MDA-MB-231转移的分子机制。方法以人乳腺癌细胞MDA-MB-231为研究材料,用小RNA干扰技术下调乳腺癌细胞HOTAIR表达,实时荧光定量聚合酶链反应检测HOTAIR下调水平。用细胞计数法检测HOTAI下调前后细胞增值能力变化,划痕实验( Wound Healing模型)检测下调HOTAIR前后乳腺癌细胞迁移能力变化。结果小干扰RNA下调HOTAIR表达后,乳腺癌MDA-MB-231细胞增殖能力显著降低( P <0.05);小干扰 RNA 下调 HOTAIR 表达后, si -HOTAIR乳腺癌细胞的迁移能力显著下降。结论长链非编码RNA HOTAIR可促进乳腺癌MDA-MB-231细胞增殖迁移,可作为乳腺癌药物研制的潜在靶点。

목적:탐토동원합기인전록반의RNA( HOTAIR)조공유선암세포MDA-MB-231전이적분자궤제。방법이인유선암세포MDA-MB-231위연구재료,용소RNA간우기술하조유선암세포HOTAIR표체,실시형광정량취합매련반응검측HOTAIR하조수평。용세포계수법검측HOTAI하조전후세포증치능력변화,화흔실험( Wound Healing모형)검측하조HOTAIR전후유선암세포천이능력변화。결과소간우RNA하조HOTAIR표체후,유선암MDA-MB-231세포증식능력현저강저( P <0.05);소간우 RNA 하조 HOTAIR 표체후, si -HOTAIR유선암세포적천이능력현저하강。결론장련비편마RNA HOTAIR가촉진유선암MDA-MB-231세포증식천이,가작위유선암약물연제적잠재파점。
Objective To evaluate the biological function of HOX tran-script antisense RNA ( HOTAIR) regulate the proliferation and metastasis of breast cancer cell.Methods The MDA -MB -231 human breast cancer cell line was used as the materials.HOTAIR expression was knowck down by small interference RNA in human breast cancer cell line MDA-MB-231.After know down the expression of the HOTAIR , the cell proliferation and migration ability of breast cancer cell was tested by Wound Healing and cell count assay.Results The cell proliferation was significant inhibit when decrease the HOTAIR expression in human breast cancer cell line MDA -MB-231 ( P<0.05 ).The migration ability of breast cancer cell was also inhibit by know down the expression of HOTAIR.Conclusion Decreased expression of HOTAIR can significant inhibit the proliferation and migration of human breast cancer cell which could be a potential treatment target for human breast cancer.
