目的 分析硫酸镁对重度子痫前期(SP)产妇Toll样受体4(TLR4)表达的影响.方法 选择同期正常产妇90例为A组、采取硫酸镁治疗的SP产妇90例为B组以及未采取硫酸镁治疗的SP产妇90例为C组,均用RT-PCR与免疫组化(SP法)检测胎盘中TLR4 mRNA和TLR4蛋白的表达,用酶联接免疫吸附试验(FLISA)法检测产妇血清中TLR4蛋白的表达,对检查结果进行分组统计并分析.结果 C组TLR4 mRNA、TLR4蛋白阳性率和血清中TLR4蛋白水平均显著高于A组(t=3.358,P<0.01),B组TLR4表达未降至正常水平,但TLR4 mRNA、TLR4蛋白阳性率和血清中TLR4蛋白水平均明显低于C组(t=2.023,P<0.05).B组患者治疗后各项实验室检查指标、不良症状与体征均明显好转(x2=49.585,P<0.01).结论 硫酸镁可能通过下调TLR4表达而起到有效的控制SP的作用.

Objective To evaluate the effect of magnesium sulfate (MgSO4) on the expression of toll-like receptor 4 (TLR4) in the patients with severe preeclampsia (SP).Methods 90 normal parturients were designated as group A,90 parturients with SP treated by magnesium sulfate were designated as group B,and 90 parturients cases without the treatment by magnesium sulfate were designated as group C.TLR4 mRNA and protein in placenta were detected by RT-PCR and immunohistochemistry.TLR4 levels in serum were studied by enzyme-linked immunosorbent assay (ELISA).Results The expression of TLR4 mRNA,TLR4 protein in placenta and the levels of TLR4 in serum of Group group C were significantly higher than those of group A (t =3.358,P <0.01).The expression of TLR4 in the group B was not reduced to the normal range level,but TLR4 mRNA,the positive rate of TLR4 protein,and the levels of TLR4 in serum were significantly lower than those in the group C (t =2.023,P < 0.05).After the treatment with magnesium sulfate,the laboratory examination,adverse symptoms and signs of the parturients in group B were significantly improved (x2 =49.585,P < 0.01).Condusion Magnesium sulfate can control SP by down-regulateding the expression of TLR4.
