目的:观察和比较致敏小鼠及对照小鼠T细胞中T‐bet、GATA3两种转录因子mRNA和蛋白表达的差异及大剂量过敏原在体外的作用。方法复制致敏小鼠模型,分离脾脏树突状细胞(D C )及T细胞。反转录‐聚合酶链反应(RT‐PCR)和Western blotting法分别检测致敏小鼠和对照小鼠 T细胞转录因子T‐bet和GATA3的mRNA及蛋白表达水平。致敏小鼠脾脏DC、T细胞与10 g/L OVA或生理盐水共培养后,RT‐PCR和Western blotting法分别检测 T‐bet和GATA3的 mRNA及蛋白表达。结果致敏小鼠T细胞T‐bet的mRNA及蛋白表达较正常对照小鼠显著降低,而GATA3的表达较对照小鼠显著升高。大剂量过敏原(10 g/L的OVA)作用下致敏小鼠T 细胞T‐bet的mRNA和蛋白表达水平较对照组显著降低,GATA3的mRNA和蛋白表达水平较对照组显著降低。结论致敏小鼠 T细胞转录因子 T‐bet表达减少而GATA3表达增加,大剂量过敏原在体外诱导致敏小鼠 T 细胞 T‐bet和GATA3表达减少,其机制可能与诱导调节性T细胞极化等有关。
目的:觀察和比較緻敏小鼠及對照小鼠T細胞中T‐bet、GATA3兩種轉錄因子mRNA和蛋白錶達的差異及大劑量過敏原在體外的作用。方法複製緻敏小鼠模型,分離脾髒樹突狀細胞(D C )及T細胞。反轉錄‐聚閤酶鏈反應(RT‐PCR)和Western blotting法分彆檢測緻敏小鼠和對照小鼠 T細胞轉錄因子T‐bet和GATA3的mRNA及蛋白錶達水平。緻敏小鼠脾髒DC、T細胞與10 g/L OVA或生理鹽水共培養後,RT‐PCR和Western blotting法分彆檢測 T‐bet和GATA3的 mRNA及蛋白錶達。結果緻敏小鼠T細胞T‐bet的mRNA及蛋白錶達較正常對照小鼠顯著降低,而GATA3的錶達較對照小鼠顯著升高。大劑量過敏原(10 g/L的OVA)作用下緻敏小鼠T 細胞T‐bet的mRNA和蛋白錶達水平較對照組顯著降低,GATA3的mRNA和蛋白錶達水平較對照組顯著降低。結論緻敏小鼠 T細胞轉錄因子 T‐bet錶達減少而GATA3錶達增加,大劑量過敏原在體外誘導緻敏小鼠 T 細胞 T‐bet和GATA3錶達減少,其機製可能與誘導調節性T細胞極化等有關。
목적:관찰화비교치민소서급대조소서T세포중T‐bet、GATA3량충전록인자mRNA화단백표체적차이급대제량과민원재체외적작용。방법복제치민소서모형,분리비장수돌상세포(D C )급T세포。반전록‐취합매련반응(RT‐PCR)화Western blotting법분별검측치민소서화대조소서 T세포전록인자T‐bet화GATA3적mRNA급단백표체수평。치민소서비장DC、T세포여10 g/L OVA혹생리염수공배양후,RT‐PCR화Western blotting법분별검측 T‐bet화GATA3적 mRNA급단백표체。결과치민소서T세포T‐bet적mRNA급단백표체교정상대조소서현저강저,이GATA3적표체교대조소서현저승고。대제량과민원(10 g/L적OVA)작용하치민소서T 세포T‐bet적mRNA화단백표체수평교대조조현저강저,GATA3적mRNA화단백표체수평교대조조현저강저。결론치민소서 T세포전록인자 T‐bet표체감소이GATA3표체증가,대제량과민원재체외유도치민소서 T 세포 T‐bet화GATA3표체감소,기궤제가능여유도조절성T세포겁화등유관。
Objective To observe the effect of high dose allergen on the mRNA and protein levels of T‐bet and GATA3 .Methods The mRNA and protein levels of T‐bet and GATA3 were detected by RT‐PCR and Western blotting in allergic mice and normal mice .And the effect of OVA at 10 g/L was observed .Results The mRNA and protein levels of T‐bet were lower in allergic mice than normal mice . And the mRNA and protein levels of GATA3 were higher in allergic mice than in normal mice .The mRNA and protein levels of T‐bet and GATA3 were significantly decreased in T cells when treated by OVA at 10 g/L in allergic mice .Conclusions The expression of T‐bet and GATA3 were suppressed by high dose allergen in allergic mice in vitro .And the underlying mechanism may be related to the regulatory T cells .
