目的:观察不同正畸力作用不同时间后,大鼠移动牙张力侧牙周组织中的BMP-2表达变化及牙周组织的改建. 方法:将健康雄性Wistar大鼠60只,随机分成4组(n=15),分别施加0 g(A组)、30 g(B组)、50 g( C组)、80 g( D组)力,建立左上第一磨牙移动模型. 分别于加力后1、3、7、14、21 d,测量各组牙齿移动的距离;取实验牙———牙周组织联合切片,HE染色法观察牙周组织变化,免疫组化法检测张力侧牙周组织中BMP-2的表达. 结果:牙齿移动以C组最大, B组次之, D组最小;HE染色显示, A组牙周组织无明显变化, B组和C组牙周组织改建活跃,张力侧可见新生的成骨细胞,D组牙周膜纤维排列混乱,并可见牙根局限性的吸收;免疫组化染色显示,BMP-2主要分布于牙周韧带中,尤其是近牙槽骨和牙骨质的区域染色较强,各加力组BMP-2表达水平均明显升高,并随着力值的增大而增加(P﹤0. 05),C组与D组相比,各时间点均无统计学差异(P>0. 05). 结论:BMP-2的表达与力值大小呈正相关,50 g的正畸力值促进牙周组织改建最明显.

AIM:To investigate the effect of different orthodontic force on BMP-2 expression in the tension side of periodontal tissues of rat during orthodontic tooth movement. METHODS:60 healthy rats were randomly divided into 4 groups (n=15) and were subjected to different orthodontic forces:group A (0 g), group B (30 g), group C (50 g) and group D (80 g). The distance of orthodontic tooth movement was determined after adding the forces for 1, 3, 7, 14 and 21 d, respectively. Then the rats were executed and the periodontal tissue of the maxiallary first molars were ob-served by HE staining. The expression of BMP-2 in periodontal tissue in the tension side was determined by immunohis-tochemical method. RESULTS:The distances of orthodontic tooth movement were as in following order:group C>B>D>A. HE staining r showed that there were no significant changes in the periodontal tissue in group A. The periodontal tissue rebuilt in groups B and C was active and osteoblasts were more in the tension side of the periodontal tissue. While in group D, periodontal ligament fibers were in disorder and root resorption was observed . Immunohistochemical staining showed that BMP-2 was mainly distributed in periodontal ligament, especially in alveolar bone and cementum region, With the increase of orthodontic force, the expression of BMP-2 increased (P ﹤0. 05). No statistically significant difference was observed between group C and group D. CONCLUSION:The expression of BMP-2 is highly related with the orthodontic force strength. 50 g orthodontic force can promote periodontal tissue remodeling.
