目的:探讨淫羊藿苷对人牙髓干细胞( hDPSCs)增殖及骨向分化的影响. 方法:MTT检测10 -2 ~10 -10 mol/L淫羊藿苷对hDPSCs增殖能力的影响,并筛选出最佳浓度(10 -5 mol/L);然后以10 -5 mol/L的淫羊藿苷作用于hDPSCs,并对其进行矿化诱导. 诱导培养21 d后,用Real-time PCR检测hDPSCs中ALP、DMP-1、DSPP各矿化相关基因的表达水平;茜素红染色观察hDPSCs矿化结节的能力. 结果:与对照组相比, 10 -2 ~10 -3 mol/L 的淫羊藿苷对hDPSCs增殖有一定的抑制作用(P<0. 05),而10 -5 mol/L和10 -6 mol/L的淫羊藿苷均能明显促进hDPSCs的增殖(P<0. 05),其中以10 -5 mol/L最好;Real-time PCR检测及茜素红染色观察结果显示,10 -5 mol/L淫羊藿苷可明显促进hDPSCs中各矿化相关基因的表达和矿化结节的形成. 结论: 一定浓度范围的淫羊藿苷可促进hDPSCs的增殖和骨向分化.

AIM:To explore the potential role of icariin in the regulation of proliferation and osteogenic dif-ferentiation of human dental pulp stem cells ( hDPSCs ) . METHODS: hDPSCs were treated by icariin at 10 -2 mol/L~10 -10 mol/L respectively. MTT assay was employed to evaluate the proliferative effect of icariin on hDP-SCs. Real time-PCR and Alizarin Red S were performed to examine the impact of icariin on osteogenic differentiation of hDPSCs. RESULTS:10 -2 mol/L~10 -3 mol/L icariin inhibited the proliferation of hDPSCs. 10 -5 mol/L icariin promoted the proliferation of hDPSCs. In addition, 10 -5 mol/L icariin could up-regulate osteogenic differentiation re-lated genes and could promote mineralized nodule formation. CONCLUSION: Icariin can enhance proliferation and osteogenic differentiation of hDPSCs at a certain concentration range.
