本研究以52头中国信阳水牛血样为材料,提取基因组DNA,采用聚合酶链反应-单链构象多态性技术(PCR-SSCP),对信阳水牛的生长素释放激素(GHRH)基因中第4外显子的突变情况进行检测。经SSCP检测发现,不同个体的PCR反应产物银染后有两种不同的带型,分别命名为AA型和AB型。针对不同带型所对应的个体PCR产物进行测序,结果表明,在该基因(GI:194719389)的第4外显子70bp处发现A→G突变,A→G导致Thr转化为Ala,在该基因的第4内含子6bp处发现A→C突变。

This study was aimed to detect SNPs of GHRH gene on 4th Exon Sequences gene in 52 blood samples from Xinyang buffalo,using the method of PCR-SSCP to extract DNA.The results from PCR-SSCP showed that there were two different band type in different after PCR reaction products silver staining of different individuals,named as AA and AB types respectively.The results from sequencing showed that A→G mutation which caused Thr trans?ferred to Ala at 70 in exon4 of the gene(GI:194719389)and A→C mutation at 6 in intron4 were detected.
