目的:建立液质联用法测定大鼠血浆中马钱苷浓度的方法,研究健康大鼠灌胃六味地黄丸后马钱苷的药代动力学特征。方法6只健康雄性 Wistar 大鼠,分别按10 g·kg -1灌胃给予六味地黄丸混悬液,按设定时间点采集血液样本,采用已建立液质联用法测定血浆中马钱苷浓度。采用 DAS 2.0药代动力学软件计算药代动力学参数。结果马钱苷主要药代动力学参数分别为 t1/2(4.398±1.633)h,Tmax(2.083±1.021)h,Cmax(369.314±53.452)ng·mL -1,AUC0~24(2275.174±553.825)ng·mL -1·h,AUC0~∞(2343.478±601.313)ng·mL -1·h。结论本研究建立的测定大鼠血浆中马钱苷浓度的液质联用法灵敏度高,重现性好,适于大鼠体内马钱苷血浓度测定及药代动力学研究。大鼠灌胃给予六味地黄丸后,马钱苷药时曲线呈双峰。

Objective To establish an HPLC - MS methods for the determination of loganin in rats plasma and to study the pharmacokinetic of loganin after administration of Liuwei Dihuang Pills. Methods 6 Wistar rats were administrated with Liuwei Dihuang Pills suspension(10 g·kg - 1 ). The blood samples were collected as scheduled time and analyzed by HPLC - MS. The pharmacokinetic parameters of loganin were calculated by DAS 2. 0 pharmacokinetic software. Results The main parameters of loganin in Liuwei Dihuang Pills were shown as follows:t1 / 2(4. 398 ± 1. 633)h,Tmax(2. 083 ± 1. 021)h,Cmax(369. 314 ± 53. 452)ng·mL - 1 ,AUC0 ~ 24(2 275. 174 ± 553. 825)ng·mL - 1 ·h,AUC0 ~ ∞(2 343 ± 601. 313)ng·mL - 1 ·h. Conclusion The HPLC - MS method was sensitive and selective for determination of loganin in plasma. A bimodal phenomenon was observed in the plasma prole after administration of Liuwei Dihuang Pills.
