目的:对比不同特异性组织来源的间充质干细胞的牙周再生能力. 方法:体外培养人牙周膜干细胞( human periodon-tai iigament stem ceiis,hPDLSCs),人颌骨来源的骨髓间充质干细胞(human jaw-derived bone marrow-derived mesenchymai stem ceiis,hJBMMSCs)和人髂骨来源的骨髓间充质干细胞(human iiiac-derived BMMSCs,hIBMMSCs),成骨诱导后进行成骨指标检测. 间接共培养JBMMSCs/IBMMSCs与PDLSCs,通过RT-PCR检测成骨相关基因,观察组织特异性来源的细胞间交互作用.构建JBMMSCs/IBMMSCs+PDLSCs细胞聚合体(ceii aggregates,CAs),检测成骨基因表达以及裸鼠皮下8 w异位牙周再生能力. 结果:经成骨诱导,hJBMMSCs的成骨相关指标表达均显著高于hIBMMSCs和hPDLSCs(P<0.05). 经JBMMSCs诱导的PDLSCs的成骨基因表达显著高于IBMMSCs(P<0.05);JBMMSCs+PDLSCs CAs成骨相关基因表达高于IBMMSCs+PDLSCs CAs(P<0.05),并且在裸鼠皮下异位再生牙周组织,前者形成排列规律的垂直于CBB表面的牙周膜样纤维,并且有更多新生骨基质沉淀. 结论:PDLSCs和JBMMSCs是具有组织特异性的间充质干细胞,是更合适牙周组织再生的种子细胞.

목적:대비불동특이성조직래원적간충질간세포적아주재생능력. 방법:체외배양인아주막간세포( human periodon-tai iigament stem ceiis,hPDLSCs),인합골래원적골수간충질간세포(human jaw-derived bone marrow-derived mesenchymai stem ceiis,hJBMMSCs)화인가골래원적골수간충질간세포(human iiiac-derived BMMSCs,hIBMMSCs),성골유도후진행성골지표검측. 간접공배양JBMMSCs/IBMMSCs여PDLSCs,통과RT-PCR검측성골상관기인,관찰조직특이성래원적세포간교호작용.구건JBMMSCs/IBMMSCs+PDLSCs세포취합체(ceii aggregates,CAs),검측성골기인표체이급라서피하8 w이위아주재생능력. 결과:경성골유도,hJBMMSCs적성골상관지표표체균현저고우hIBMMSCs화hPDLSCs(P<0.05). 경JBMMSCs유도적PDLSCs적성골기인표체현저고우IBMMSCs(P<0.05);JBMMSCs+PDLSCs CAs성골상관기인표체고우IBMMSCs+PDLSCs CAs(P<0.05),병차재라서피하이위재생아주조직,전자형성배렬규률적수직우CBB표면적아주막양섬유,병차유경다신생골기질침정. 결론:PDLSCs화JBMMSCs시구유조직특이성적간충질간세포,시경합괄아주조직재생적충자세포.
Objective:To compare the periodontai regeneration of different specific-tissue derived mesenchymai stem ceiis .Meth-ods:Human periodontai iigament stem ceiis ( hPDLSCs ) , human jaw-derived bone marrow-derived mesenchymai stem ceiis (hJBMMSCs) and human iiiac-derived bone marrow-derived mesenchymai stem ceiis(hJBMMSCs) were cuitured in vitro.After osteo-genic induction ,reiative osteogenic tests were performed .PDLSCs induced by JBMMSCs/IBMMSCs were tested for osteogenic expression by RTPCR.Ceii aggregates (CAs) of PDLSCs and JBMMSCs/IBMMSCs were compound.The osteogenic genes expression were tested;PDL regeneration in nude mice ectopic transpiantation was observed .Results:After osteogenic induction , reiative osteogenic tests of JBMMSCs were higher than those of IBMMSCs and PDLSCs .PDLSCs induced by JBMMSCs expressed stronger osteogenic genes than those by IBMMSCs .CAs of PDLSCs/JBMMSCs expressed stronger osteogenic genes ,made more reguiar and verticai inserting fibers into the surface of CBB,regenerated more new bone matrix than CAs of PDLSCs /IBMMSCs.Conclusions:JBMMSCs and PDLSCs are spe-cific-tissue MSCs and more appropriate for PDL regeneration .
