目的:检测骨肉瘤活检标本中COX‐2与Survivin蛋白的表达水平,并分析其与骨肉瘤临床病理特征之间关系。方法:收集新鲜切除并经临床病理学确诊的骨肉瘤标本30例、骨软骨瘤标本20例。其中骨肉瘤标本按临床分期标准,Ⅰ期7例,Ⅱ期14例,Ⅲ期9例。采用免疫组织化学方法和Western blot法检测骨肉瘤组织、骨软骨瘤组织及正常组织中COX‐2及Survivin蛋白表达。结果:Survivin 和COX‐2蛋白主要表达于细胞质中,在骨肉瘤和骨软骨瘤组织中的表达分别为63.3%和73.3%、30.0%和25.0%;Spearman等级相关分析表明,在骨肉瘤组织中,Survivin与COX‐2蛋白表达水平为正相关关系(r=0.775,P<0.05)。Western blot结果显示,骨肉瘤组织中Survivin和COX‐2蛋白表达水平显著高于骨软骨瘤组织及正常组织组(P<0.05);SPSS13.0统计软件分析结果,未见Survivin和COX‐2蛋白表达与骨肉瘤年龄、病理分级和类型之间存在显著相关性。结论:Survivin与COX‐2蛋白在骨肉瘤组织中高表达,且二者呈正相关关系,表明Sur‐vivin与COX‐2可能在骨肉瘤的发生发展中起重要作用。

목적:검측골육류활검표본중COX‐2여Survivin단백적표체수평,병분석기여골육류림상병리특정지간관계。방법:수집신선절제병경림상병이학학진적골육류표본30례、골연골류표본20례。기중골육류표본안림상분기표준,Ⅰ기7례,Ⅱ기14례,Ⅲ기9례。채용면역조직화학방법화Western blot법검측골육류조직、골연골류조직급정상조직중COX‐2급Survivin단백표체。결과:Survivin 화COX‐2단백주요표체우세포질중,재골육류화골연골류조직중적표체분별위63.3%화73.3%、30.0%화25.0%;Spearman등급상관분석표명,재골육류조직중,Survivin여COX‐2단백표체수평위정상관관계(r=0.775,P<0.05)。Western blot결과현시,골육류조직중Survivin화COX‐2단백표체수평현저고우골연골류조직급정상조직조(P<0.05);SPSS13.0통계연건분석결과,미견Survivin화COX‐2단백표체여골육류년령、병리분급화류형지간존재현저상관성。결론:Survivin여COX‐2단백재골육류조직중고표체,차이자정정상관관계,표명Sur‐vivin여COX‐2가능재골육류적발생발전중기중요작용。
Objective :To investigate the expression of COX‐2 and survivin and clinical significance in hu‐man osteosarcomas ,and explore their correlation with the pathologic characters in patients with osteosarcomas tis‐sue .Methods :Fifty freshly resected OS and OC samples were collected at the orthopedics department of three hospi‐tals and were classified according to American Joint Committee on Cancer TNM staging system .There were 30 cases of OS tissues and 20 cases of OC tissues .All tumors and normal tissues were diagnosed by two independent gastro‐enterologists .The expression of COX‐2 and survivin was assessed using immunohistochemical assay and Western blot assay in biopsy samples .Results :The IHC assays shown that the expression of COX‐2 and survivin was mainly localized in the cytoplasm .The expression of COX‐2 and survivin was respectively observed in 73 .3% and 25 .0%OS tissues and in 63 .3% and 30 .0% OC tissues ,indicating their higher expression in OS compared with OC . Spearman rank correlation analysis showed a positive correlation between COX‐2 and survivin expression in OS (r=0 .775 ,P<0 .05) .The relationship between the expression of COX‐2 and survivin and clinicopathologic features was analyzed .No significant correlation was found between the expression of COX‐2 and survivin with age and pathologi‐cal grade and types of OS .Conclusion :The higher expression of COX‐2 and survivin in osteosarcoma and positive correlation between COX‐2 and survivin suggesting that they may be provide a potential therapeutic strategy for the treatment in osteosarcoma .
